摘要
目的探索慢性HBV感染免疫耐受期及免疫清除期肝组织微小RNA(miRNA)表达谱的差异并探讨其临床意义。方法应用miRNA芯片筛选9例HBV感染免疫耐受期、8例HBV感染免疫清除期及6例健康对照者肝组织miRNA分子表达谱,进行组间两两比较后,筛选出肝组织中差异表达的miRNA。结果与健康对照组相比,慢性HBV感染免疫耐受期肝组织中miRNA分子上调9个,下调6个;慢性HBV感染免疫清除期肝组织中miRNA分子上调8个,下调6个。与慢性HBV感染免疫耐受期相比,免疫清除期肝组织中miRNA分子上调6个,下调1个。结论慢性HBV感染免疫耐受期和免疫清除期肝组织差异表达的miRNA可能与HBV感染后的免疫清除过程密切相关,有可能成为临床鉴别慢性HBV感染免疫耐受和免疫清除状态的新型分子标记。
Objective To explore the difference in expression profiles and their clinical significance of microRNA molecules in the liver between immune tolerance and immune clearance phase of chronic hepatitis B vius (HBV) infection . Methods The miRNA molecules expressions of 9 immune‐tolerance patients ,8 immune‐clearance patients of chronic HBV infection ,and 6 healthy controls were screened using miRNA microarray . The difference in expression profiles of microRNA molecules in the liver was screened out through comparison among groups .Results Compared with the healthy controls ,significant differences were found in the expression profiles of miRNA molecules in the liver in immune tolerance phase (9 molecules up‐regulated ,and 6 down‐regulated) and immune clearance phase(8 molecules up‐regulated ,and 6 down‐regulated) of chronic HBV infection .Compared with immune tolerance phase ,6 molecules up‐regulated and 1 down‐regulated in the immune clearance phase .Conclusion The abnormal expression profiles of miRNAs in the liver between immune tolerance phase and immune clearance phase of chronic HBV infection could be closely associated with immune clearance during chronic HBV infection and may be new molecular markers to distinguish immune clearance from immune tolerance phase of chronic HBV infection in the clinical context .
