摘要
目的:探讨不同浓度的溶血卵磷脂(LPC)对人视网膜微血管内皮细胞表达糖基化终产物受体的影响。方法:人视网膜内皮细胞加入不同浓度的LPC:(20μmol/L,40μmol/L,60μmol/L)和糖基化终产物,培养24 h,Real-time(RT)-PCR检测糖基化终末产物受体(RAGE)m RNA表达水平,Western blot检测RAGE蛋白表达水平。结果:Realtime(RT)-PCR及Western blot结果表明,LPC的溶度为60 umol/L作用24 h后,RAGE的m RNA及蛋白表达水平明显高于溶度为20μmol/L、40μmol/L。结论:LPC能促进人视网膜内皮细胞RAGE的表达,并且呈剂量依赖性。
Objective The aim of the present study was to investigate the effect of LPC on expression of glycation end products receptor(RAGE) in human retinal endothial cells. Methods HRECs were treated or not treated with AGEs in the presence or absence of LPC, The expression of RAGE were assessed by Western blot analysis using specific antibodies and Real Time-PCR. Results Real-time-PCR and western blot results showed that RAGE expression levels were significantly higher with treatment of LPC 60 umol/L for 24 hours than LPC 20 umol/L, 40 umol/L. Conclusion These results demonstrate that treatment of HRECs with LPC caused dose- dependent increases in RAGE expression.
