莱菔硫烷对H446细胞株的抑制作用及CYR61的影响

摘要 目的探讨莱菔硫烷(sulforaphane,SFN)对小细胞肺癌H446细胞株的抑制作用及CYR61基因表达的影响。方法 MTT法检测SFN对H446细胞的抑制作用;流式细胞仪检测细胞凋亡情况;Real-time PCR检测H446肺癌细胞株CYR61表达。结果 MTT结果显示:SFN从20μM的浓度起开始出现细胞抑制效应,抑制率与SFN浓度呈正相关关系。40μM SFN干预组的CYR61基因表达量较高,与其他组之间差异有统计学意义。结论 SFN在一定浓度下对H446细胞株有抑制作用,并促进CYR61基因表达,为食用十字花科类蔬菜防治肺癌提供依据。 Objective To explore the inhibition of SFN on H446 cell lines and its effects on CYR61 expression. Methods The inhibition of SFN on H446 cell lines was detected by MTT. The cell apoptosis was detected by flow cytometry. CYR61 expression level in H446 lung cancer cell lines was identified by real-time PCR. Results MTT results indicated that SFN had a restrain effect with a dose-dependent manner when the concentration exceeded 20 μM. The expression of CYR61 was higher in the SFN intervention group of 40 μM and its difference compared with other groups has statistical significance. Conclusion SFN inhibited the growth of H446 cells in a concentration-depended manner and promoted CYR61 expression of H446 lung cancer cell lines,which provides basis for vegetable-broccoli in the prevention of lung cancer.
