摘要
目的探讨针刺对血管性痴呆大鼠血清中Bax和Bcl-2表达的影响。方法将大鼠随机分为假手术组、模型组和针刺组。间断夹闭颈总动脉法建立Va D模型,Morris水迷宫实验检测模型是否成功,酶联免疫吸附法分析(ELISA)检测血清中Bax和Bcl-2的含量。结果针刺治疗后Bax表达较模型组〔(410.176±65.942)pg/m L比(612.353±92.554)pg/m L〕下降(P<0.01),Bcl-2较模型组表达〔(275.509±60.833)pg/m L比(119.246±20.562)pg/m L〕明显升高(P<0.01)。结论针刺可通过上调大鼠Bcl-2水平,下调Bax表达,对抗脑缺血所致的细胞凋亡,防止继发性损伤,达到促进神经修复的作用。
Objective To investigate the role of needling affect to expression of Bax and Bcl-2 of the rats after vascular dementia( Va D). Methods The rats were randomly divided into sham operation group,Va D model group and Needling treatment group. The rat model of Va D was built by intermittently clipping common carotid artery. Morris water maze test whether the surgery had been successful and detection the changes of Bax and Bcl-2 by Enzyme-linked immunosorbent assay( ELISA). Results Bax expression( 410. 176 ± 65. 942) pg / m L compare( 612. 353 ± 92. 554)pg / m L decreased after acupuncture treatment( P〈 0. 01),the Bcl-2 expression( 275. 509 ± 60. 833) pg / m L compares( 119. 246 ± 20. 562) pg / m L increased significantly,Bax / Bcl-2 ratio decreased compared with model group( P 〈0. 01). Conclusion The acupuncture therapy on vascular dementia maybe related to up-regulated the expression of Bcl-2 protein in rat and decreased Bax protein expression thus regulate anti-apoptotic effect and promotes neurological function recovery in cerebral ischemia injury and prevent secondary injury,to promote the role of nerve repair.
