摘要
目的 了解胚胎培养液添加白蛋白对pH的影响.方法 在HTF受精液、CM卵裂液、G1卵裂液和CSC全程培养液中添加不同浓度的白蛋白制剂,在经过5.0% CO2过夜平衡后用血气分析仪检测其HCO3-浓度和pH.结果 白蛋白制剂的HCO3-浓度明显低于培养液;培养液中添加白蛋白会降低HCO3-浓度和pH,白蛋白浓度与pH呈高度负相关(P<0.01);不同品牌培养液的pH,模型存在较明显差异,HTF和CM的pH明显低于G1和CSC (P<0.01).结论 胚胎培养液中添加白蛋白会降低pH且与培养液的品牌相关,应通过实测胚胎培养液的pH来确定CO2浓度,尤其是搭配使用不同品牌产品的时候,仅仅依靠经验或厂家提供的参数是不可靠的.
Objective To explore the effect of albumin on the pH value of embryo culture medium. Methods Firstly, albumin with different concentrations were added into HTF, CM, G1 and CSC culture medium. Then these cultures were incubated in incubator with CO2 of 5.0% overnight. Lastly, the concentration of HCO3 and pH value were detected using blood-gas analyzer. Results The concentration of HCO3- of albumin was significantly lower than that of cultures. Adding albumin into these cultures can reduce the concentration of HCO3 and pH value, and the concentration of albumin was highly negatively correlated with pH value (P〈0.01). The pH value of different culture was different, and the pH value of HTF and CM was significantly lower than that of G 1 and CSC (P〈0.01). Conclusion Adding albumin into culture medium can reduce it's pH value, which may be related to different products of culture medium.It's not reliable to determine the concentration of CO2 only based on experience or parameters provided by manufacturers, especially when using different culture oroduct together. We should actually detect oH value of the cultures.
