CK15在食管鳞癌患者组织及血清中的表达与意义 被引量:1

摘要 目的探讨细胞角蛋白15(cytokeratin15,CK15)在食管鳞癌患者组织及血清中的表达及其意义,为食管鳞癌的诊断提供依据。方法 1取30例食管鳞癌患者癌组织及癌旁组织应用Western blot法检测CK15在两组中的表达。2采集食管鳞癌患者组及体检的正常对照组受检者血清,应用ELISA法检测两组血清中CK15、增殖细胞核抗原(PCNA)的表达,应用化学发光法检测两组血清中细胞角蛋白19片段抗原(CYFRA21-1)的表达。结果 1与癌旁组织比较,CK15在食管鳞癌组织中的表达明显减少(P<0.05)。2与正常对照组血清比较,CK15在食管鳞癌患者组血清中的表达明显降低(P<0.05);PCNA在两组血清中表达无明显差异(P>0.05);CYFRA21-1在食管鳞癌患者组血清中的表达明显升高(P<0.05)。3单项检测血清中CK15、CYFRA21-1水平对食管鳞癌诊断的敏感性分别为33.33%和30.00%,而联合检测敏感性为60.00%。结论CK15在食管鳞癌患者组织与血清中的表达下调可能与食管鳞癌的发生、发展有关。联合检测CK15、CYFRA21-1对食管鳞癌的诊断有重要价值。 Objective To explore the expression of cytokeratin 15 (CK15) in the tissues and serum of the patients with esophageal squamous cell carcinoma(ESCC) and its significance,and to provide evidences for the diagnosis of ESCC.Methods ①Western blot was used to detect the expression of CK15 in 30 cases of esophageal cancer tissues and para-carcinoma esophageal tissues.②Serum levels of CK15 and PCNA were detected by ELISA in 30 cases of esophageal squamous cell carcinoma and 30 normal controls.Serum level of cytokeratin 19 fragment antigen(CYFRA21-1) was detected by chemiluminescence in two groups.Results ①The expression of CK15 was significantly lower in esophageal cancer tissues than that in para-carcinoma esophageal tissues(P < 0.05).②Compared with control group,the expression of CK15 in serum was significantly reduced in esophageal cancer patients(P < 0.05).Expression of PCNA in the serum of patients with esophageal carcinoma and the normal controls showed no difference (P > 0.05).Expression of CYFRA21-1 in the serum of esophageal carcinoma was significantly higher than that of controls(P < 0.05).③The sensitivities of CK15,CYFRA2l-1 and their combination in patients with esophageal squamous cell cancer were 33.33%,30.00% and 60.00%.Conclusion The lower expression of CK15 in tissues and serum is closely related to the occurrence and development of esophageal squamous cell carcinoma.The combined detection of CKI5 and CYFRA21-1 is of great value in the diagnosis of esophageal squamous carcinoma.
