Anesthetic Effects of 3-Aminobenzoic Acid Ethyl Ester Methanesulfonate(MS-222) on Misgurnus anguillicaudatus

摘要 根据泥鳅(Misgurnus anguillicaudatus)对20 - 140 mg/L 麻醉剂间氨基苯甲酸乙酯甲磺酸盐(MS-222)在最终麻醉状态和复苏过程中的行为特征,分别把麻醉程度和复苏过程分为6 和4 个时期,研究140 - 340 mg/L的麻醉剂MS-222 在12 ℃和22 ℃条件下对泥鳅的麻醉效果,比较180 mg/L 的MS-222 对不同规格(5.1、11.6、28.1 g)泥鳅的麻醉效果差异.结果表明:当MS-222 溶液浓度为80 mg/L 时,泥鳅达到麻醉3 期,可以作为泥鳅长途运输的参考剂量;泥鳅呼吸频率受麻醉程度的影响,在麻醉3 期之前无显著变化,但是4 期之后出现显著下降(P 〈 0.05);在MS-222 质量浓度为140 - 340 mg/L 时,水温12 ℃条件下的鱼体至麻醉4 期的时间(下称入麻时间)小于22 ℃,水温12 ℃条件下麻醉6 期的鱼体至复苏2 期的时间(下称复苏时间)亦小于22 ℃;在水温12 ℃和22 ℃时,MS-222 对泥鳅有效麻醉的质量浓度分别为180 mg/L 和220 mg/L;体质量小的泥鳅入麻时间短,复苏时间长(P 〈 0.05).MS-222 对泥鳅有较好的麻醉效果,在麻醉操作时,需综合考虑水温、个体规格等因素,使用合理的麻醉浓度. Based on the stress reaction of the loach(Misgurnus anguillicaudatus) behavior characteristics and respiratory frequency at 20- 140 mg/L concentrations by 3-Aminobenzoic acid ethyl ester methanesulfonate(MS-222), the final anesthetic status can be divided into six stages, and the revival process into four stages. The anesthetic effects of MS-222 on loach was studied under the conditions of different MS-222 concentrations(140- 340 mg/L) and water temperature(12 and 22 ℃), and different weights(5.1, 11.6 and 28.1 g) of the loach at 180 mg/L concentration of MS-222. The results showed that the loach can reach anesthesia stage 3 at 80 mg/L of MS-222 solution concentration, and it can be a reference concentration for loach long-distance transportation; The breath frequency of loach showed no significant changes when the loach were in the final anesthetic stages 1 to 3, and declined clearly until stage 4 under the concentration range of 20- 140 mg/L. The induction time of the loach under the condition of temperature 12 ℃,in the MS-222 solution with mass concentration of 140- 340 mg/L, were longer than those under the condition of temperature 22 ℃, and the change of recovery time was same to induction time. The effective anesthetic concentrations of MS-222 on loach at 12 and 22 ℃ are 180, 220 mg/L, respectively. Small loach individuals have short anesthesia time and long recovery time(P < 0.05). It is suggested that MS-222 has preferable anesthetic effect to the loach, and at the anesthesia operation, we need to consider factors such as water temperature, fish size in order to use reasonable anesthesia concentration for the loach.
