女贞花总黄酮测定及对亚硝酸盐清除作用研究

Study on Determination and Scavenging Action of Total Flavonoids in Ligustrum Flowers

利用分光光度法测定了女贞花中总黄酮含量,研究了其对亚硝酸盐的清除作用。结果表明：对于女贞花中总黄酮测定的最佳显色体系为Al（NO3）3-NaNO2-NaOH显色体系,最佳测定时间为显色后20～50min范围内,测定方法精密度RSD=0.37%（n=7）,回收率102.03%（RSD=0.88%）,利用该法测得女贞花样品总黄酮含量为14.28%（RSD=4.01%,n=5）;在体外模拟胃液条件下,提取液总黄酮浓度在4～96μg/mL范围内,清除率从（11.84±0.56）%增加到（87.21±1.29）%,半数清除率IC50值为20.4μg/mL。该测定方法简便准确,女贞花提取液对亚硝酸盐的清除效果显著。

In this work,a study was made on the content and scavenging action of flavonoids in Ligustrum Flowers on nitrite.The content of total flavonoids was determined using Al（NO3）3-NaNO2-NaOH chromogenic system and spectrophotometry.The results showed that the optimal time to scan the absorption spectrum was 20min≤t≤ 50 min,the relative standard deviation（RSD）of precision test was 0.37%（n=7）,the mean recovery rate was 102.03%（RSD=0.88%）and total flavonoids yield in Ligustrum Flowers sample was 14.28%（RSD=4.01%,n=5）.Under simulated human gastric juice environment,the clearing rate increased from（11.84±0.56）% to（87.21±1.29）% when the average of flavonoids was from 4μg/mL to 96μg/mL.The content of total flavonoids and its function of scavenging nitrite in Ligustrum Flowers were discussed accurately.
