肺癌细胞株A549、H1650、DMS53中整合素α3的表达比较 被引量:2

摘要 目的比较整合素α3(ITGA3)在三种肺癌细胞株A549(肺腺癌)、H1650(肺泡细胞癌)、DMS53(小细胞肺癌)中的表达。方法应用基因芯片技术提取A549、H1650、DMS53细胞株的mRNA,反转录为c DNA,以32P标记c DNA,与整合素基因芯片进行杂交,经放射自显影后读取灰度值,并以基因表达的限制性分析(RAGE)和流式细胞仪技术进行验证。结果 ITGA3亚单位在A549、H1650、DMS53细胞株中的表达分别为7.58±1.37、8.10±1.51、2.38±0.20,A549、H1650与DMS53细胞株比较,P均<0.01。RAGE结果显示,经AvaⅡ酶切后,A549、H1650、DMS53细胞株均出现ITGA3(69 bp),分别为167.0、172.1、44.7,A549、H1650与DMS53细胞株比较,P均<0.01。流式细胞仪检测结果显示,ITGA3在A549、H1650、DMS53细胞株中的表达分别为83.62±1.40、90.86±4.78、1.24±0.53,A549、H1650与DMS53细胞株比较,P均<0.01。结论 ITGA3在A549和H1650细胞株中的表达上调,高于其在DMS53细胞株中的表达。
