摘要
目的观察肺癌发生过程中基质金属蛋白酶(MMP-9)和上皮性钙黏附素(E-cadherin)的表达,探讨两者临床应用价值。方法收集我院肺癌患者肺组织石蜡标本78例、良性肺组织石蜡标本28例及正常肺组织石蜡标本10例,采用流式细胞免疫法定量检测肺组织MMP-9和E-cadherin的表达。结果肺癌组中的MMP-9的表达显著高于良性对照组,Ⅲ、Ⅳ期肺癌组织中MMP-9的表达显著高于Ⅰ、Ⅱ期,Ⅲ级(低分化)肺癌组织中的MMP-9的表达高于Ⅰ级、Ⅱ级(高中分化)肺癌组织,其差异均具有有统计学意义(P<0.05)。肺癌组中的E-cadherin显著低于良性对照组,Ⅲ、Ⅳ期肺癌组织中E-cadherin的表达显著低于Ⅰ、Ⅱ期,Ⅲ级(低分化)肺癌组织中的E-cadherin的表达低于I级、Ⅱ级(高中分化)肺癌组织,其差异均具有统计学意义(P<0.05)。结论肺组织中MMP-9和E-cadherin可能参与了肺癌的发生、发展,联合检测可以作为肺癌治疗、预后的判断指标。
Objective To investigate the expression of matrix metalloproteinase 9 (MMP-9) and epithelial cadherin (E-cadherin) in lung cancer and their clinical value. Methods 78 cases of paraffin specimens were collected from lung cancer tissue, 28 cases of benign lung tissues and 10 cases of normal lung tissues. Flow cytometry immune method was used to detect the expression of MMP-9 and E-cadherin. Results The expression of MMP-9 was higher in lung cancer tissue than in benign lung tissue, and it was obviously higher in the groups of stage III and IV than in the groups of stage Ⅰ and Ⅱ. The expression of MMP-9 at grade Ⅲ (low differentiation) was higher than that at grade Ⅰ andⅡ (medium and high differentiation) (P 〈 0.05 ). The expression of E-cadherin was lower in lung cancer tissue than in benign tissue, and it was obviously lower in the groups of stage Ⅲ and IV than in the groups of stage Ⅰ and Ⅱ. The expression of E-cadherin at stage Ⅲ (low differentiation) was lower than that at stage Ⅰ and Ⅱ (medium and high differentiation) (P 〈 0.05). The expression of E-cadherin had a negative correlation with the expression of MMP-9 protein ( r = - 0. 496, P 〈 0.01 ). Conclusion MMP-9 and E-cadherin in lung tissue may participate in the process of the occurrence and development of lung cancer, and the joint detection can be used as a marker in the treatment and prognosis of lung cancer.
