摘要 目的应用高效液相色谱法(HPLC)测定小柴胡汤剂中黄芩苷含量。方法小柴胡汤以水煎法制备,其中黄芩苷的含量用HPLC法进行分析,色谱柱为TSKgel ODS C18(250 mm×4.6 mm,5μm),柱温30℃,流动相为甲醇:水:磷酸(体积比为65∶35∶0.7),流速1 m L/min,检测波长280 nm。结果黄芩苷的线性范围为0.025~1.6 mg/m L,标准曲线为y=18533x-724.89,R2=0.9993,平均加样回收率为94.58%(RSD=1.76%),稳定性考察表明小柴胡汤室温放置48 h,其中黄芩苷含量稳定,小柴胡汤中黄芩苷平均含量为3.39 mg/m L(RSD=1.39%)。结论该方法简便易行、灵敏度高、准确可靠,可用于小柴胡汤的质量控制。 Objective To establish the HPLC method for determination of baicalin in Xiaochaihu decoction.Methods Xiaochaihu decoction was prepared through extracting method of decoction with the prescription according to Xiaochaihu granules in the Chinese Pharmacopoeia( 2010 edition) and then baicalin in the Xiaochaihu decoction was determined through column of TSKgel ODS C18( 250 mm × 4. 6 mm,5 μm) with column temperature of 30 ℃,mobile phase of methyl alcohol- water phosphoric acid( 65: 35: 0. 7) with a flow rate of 1. 0 ml / min and at detection wavelength of 280 nm. Results A good linearity can be seen from the calibration curve of baicalin in the range of 0. 025 mg / ml to 1. 6 mg / ml with R2 of 0. 999 3 and the average recovery was 94. 58% with RSD of 1. 76%. Xiaochaihu decoction was stable in 48 hours after preparation. The average content of baicalin in Xiaochaihu decoction was 3. 39 mg / ml( RSD = 1. 39%). Conclusion HPLC method might be ideally used to the quality control of Xiaochaihu decoction for convenient,highly sensitive,accurate and reliable characteresitic.
