积雪草苷对Aβ1-42致内皮细胞炎症因子表达的影响 被引量:1

摘要 目的探索积雪草苷对Aβ1-42诱导内皮细胞炎症因子表达影响。方法将人脐静脉血管内皮细胞株(human umbilical vein endothelial cells,HUVEC)分为模型组,阴性对照组,积雪草苷10-5、10-6、10-7、10-8mol/m L组,共6组。模型组给予50 nmol/m L Aβ1-42刺激HUVEC细胞24 h,积雪草苷各组在给予Aβ1-42刺激同时给予积雪草苷10-5、10-6、10-7、10-8mol/m L干预。CCK-8试剂盒(cell counting kit-8,CCK-8试剂盒)法检测HUVEC细胞活性;Western-blot检测COX-2蛋白表达;ELISA法检测IL-1,IL-6和TNF-α含量。结果与模型组比较,积雪草苷各浓度组可以明显提高β淀粉样蛋白刺激后血管内皮细胞活性,减少细胞COX-2蛋白表达(P<0.05),抑制β淀粉样蛋白致HUVEC细胞的IL-1,IL-6与TNF-α表达,有统计学意义(P<0.05)。结论积雪草苷对Aβ1-42致内皮细胞炎症因子表达具有保护作用。 Objective To explore the effect of asiaticoside on expression of inflammatory factors of human umbilical vein endothelial cells( HUVEC) cells induced by Aβ1-42. Methods HUVEC cell lines were divided into 6 groups: model group,control group,asiaticoside 10^-5,10^-6,10^-7,10^-8mol / m L group. Model group was stimulated with 50 nmol / m L Aβ1-42 and last 24 h; asiaticoside groups were administrated asiaticoside10^-5,10^-6,10^-7,10^-8mol / m L on the basis of Aβ1-42 stimulation and last 24 h. HUVEC viability were detected by CCK-8,expression of COX-2 protein were detected by Western blot; the concentration of IL-1,IL-6 and TNF-α were detected by ELISA. Results Compared with model group,10-5,10-6,10-7,10-8mol / m L asiaticoside could increase HUVEC cell survival rate,reduce the expression of COX-2 significantly( P〈0. 05),inhibite the concentration of IL-1,IL-6 and TNF-α,there was statistical significance( P〈0. 05). Conclusion Asiaticoside has a protective effect of HUVEC cells inducing by Aβ1-42.
