苦参碱对大鼠脑缺血再灌注损伤后脑细胞凋亡和超微结构的影响 被引量:1

Effects of matrine on ultrastructure and apoptosis of brain cell after cerebral ischemia reperfusion injury in rats

摘要 目的探讨苦参碱对大鼠脑缺血再灌注损伤的神经保护作用。方法sD大鼠60只随机分为6组:假手术对照组、手术模型组、苦参碱(4、8、16mg/kg)预处理组、依达拉奉处理组(1mg/kg)。手术前48小时,假手术对照组和手术模型组大鼠腹腔注射生理盐水,各预处理组给予相应的药物。每日2次,每次2ml,共给药2日。于脑缺血再灌注后,观察各组大鼠造模后48小时内的神经行为学评分,计算各组大鼠脑梗塞面积,流式细胞仪测定脑细胞凋亡,透射电镜观察海马区脑细胞的超微结构变化。结果假手术组神经行为学评分无明显改变,手术模型组术后48小时内的神经行为学评分高于假手术组。与模型组相比,苦参碱各预处理组和依达拉奉注射液组神经行为学评分均降低,差异有统计学意义(P〈0.05)。与假手术组比较,手术模型组脑梗塞面积明显增大,与手术模型组比较,苦参碱8、16mg/kg剂量组、依达拉奉组脑梗塞面积均明显下降,差异有统计学意义(P〈0.05)。与假手术组比较,模型组脑细胞凋亡明显升高;与模型组比较,苦参碱各预处理组、依达拉奉注射液组大鼠脑细胞凋亡明显降低,差异有统计学意义(P〈0.05)。假手术组神经元细胞无明显损伤改变,手术模型组神经元细胞细胞膜及胞内细胞器明显发生损伤改变,苦参碱各预处理组和依达拉奉注射液组神经元细胞损伤改变明显降低。结论苦参碱对大鼠脑缺血再灌注损伤具有神经细胞保护作用。 Objective To investigate the neuroprotective effect of matrine on cerebral ischemia- reperfusion injury in rats. Methods 60 SD rats were randomly divided into 6 groups, namely sham opera- tion group, model group, matrine pretreatment group ( 4,8,16 mg/kg), edaravone group ( 1 mg/kg). 48 hours before operation, rats in both sham operation group and model group were gave saline by intraperito- neal injection, other drug pretreatment group were gave matrine injection or edaravone, respectively, b. i. d for 2 days. Groups except of sham operation group were established model of cerebral ischemia-reperfusion injury in rats. 48 hours after operation, neuro-behavior was scored, cerebral infarct area was calculated, brain cell apoptosis was determined by flow cytometry and ultrastructural changes of hippocampus cells were observed by transmission. Results Neurobehavioral score of rats in sham operation group didn' t change and was lower than that of model group. Neurobehavioral score of Both matrine and edaravone pre- treatment groups were significantly lower than that of model group. Cerebral infarct area increased so as to the model building operation. But matrine (8,16 mg/kg) or edaravone pretreatment can decrease infarct area. Operation did make brain cell apoptosis and nerve injury in rats and both matrine and edaravone pre- treatment have obviously decreased them. Conclusion Matrine shows cerebral protective effect in ische- mia reperfusion injury.
