间充质干细胞移植对阿霉素肾病的肾脏保护作用 被引量:1

The protective effect of human umbilical cord mesenchymal stem cells on adriamycin nephropathy

摘要 目的探讨人脐带间充质干细胞(HUMSCs)移植对阿霉素肾病(ADR)的肾脏保护作用。方法贴壁法培养HUMSCs,将SD大鼠随机分为4组,每组6只:(1)ADR模型组;(2)HUMSCs移植(ADR+MSC)静脉低剂量治疗组,大鼠在第1天尾静脉注射HUMSCs细胞悬液2×106个;(3)ADR+MSC静脉高剂量治疗组,大鼠在第1、8、15、22天尾静脉注射HUMSCs细胞悬液2×106个,而ADR组尾静脉注射等剂量的DMEM培养液1ml;(4)对照组均给予等体积生理盐水。结果ADR+MSC组能改善阿霉素肾病的尿蛋白、降低血胱抑素c(CysC)水平,血清中自细胞介素(IL)-6及肿瘤坏死因子(TNF)-α水平明显下降。结论HUMSCs移植可改善ADR大鼠的临床及实验室检查指标,延缓ADR的进展,其机制可能是通过内分泌或旁分泌途径发挥肾脏保护作用。 Objective To evaluate stem cell therapeutic effect on adriamycin nephropathy. Methods Human umbilical cord mesenchymal stem cells (HUMSCs) were incubated by adherence. The Sprague Dawley rats were randomly assigned to four groups, no intervention was applied to the rats of nor- mal group. Model group and transplant group rats were respectively injected adriamycin 4mg/kg and 3.5 mg/kg via tail vein on dayl and day8. ADR + MSC iv Low group rats were infused with stem cells(2 × 106) via tail vein on dayl ,day8 ,dayl5 and day22. ADR + MSC iv High group rats were infused with stem cells(2 × 106) via tail vein on dayl ,day8,dayl5 and day22,whereas model group rats were infused with DMEM lml via tail vein at same time. Results Human umbilical cord mesenchymal cells administration markedly alleviated urine protein concentration,decrease serum Cys C level. Further investigation showed that human umbilical cord mesenchymal cells reduced the serum IL-6 and TNF-α level. Conclusion Transplantation of human umbilical cord mesenchymal cells can improve clinical and laboratory finding of adriamycin nephrop- athy and delay the progress of adriamycin nephropathy. Results have suggested that human umbilical cord mesenchymal cells play a protective effect in ardriamycin nephropathy rats via endocrine or paracrine way.
