抗氧化剂对砷诱导人正常膀胱上皮细胞HIF-1α表达的影响

Effect of antioxidants on arsenic-induced HIF-1α expression in human urothelial cells

摘要 目的探讨抗氧化剂对砷诱导人正常膀胱上皮细胞HIF-1α表达的影响。方法人膀胱上皮细胞(SVHUC-1)暴露于0、0.5、1、2、4、8、10μmol/L亚砷酸钠(Na As O2)24 h,RT-PCR检测HIF-1α的mRNA水平;选择4和10μmol/L剂量组,分别处理4、12、24、48、72 h,再以RT-PCR法检测HIF-1α的mRNA水平;SV-HUC-1细胞同时暴露于Na As O2和抗氧化剂N-乙酰半胱氨酸(NAC)、维生素C(VC),检测HIF-1α的蛋白含量。结果经砷处理后,SV-HUC-1细胞HIF-1α的mRNA含量显著升高(P<0.05),并与染砷剂量呈显著剂量-反应关系;抗氧化剂NAC和VC均显著降低了HIF-1α的蛋白水平。结论砷能诱导人正常膀胱上皮细胞HIF-1α表达增加,抗氧化剂NAC和VC对砷诱导的HIF-1α表达具有显著的抑制作用。 Objective To explore the effect of antioxidants to arsenic-induced HIF-1α expression in human urothelial cells.Methods SV-HUC-1 cells were treated with Na As O2( 0、0. 5、1、2、4、8、10 μmol/L,respectively) for 24 h,their mRNA expression levels of HIF-1α were analyzed using RT-PCR,β-actin as a loading control. Then SV-HUC-1 cells were treated with 4 and 10 μmol / L with Na As O2 together for 4、12、24、48 and 72 h respectively,the cells expression levels of HIF-1αwere analyzed using RT-PCR as well and the protein expression levels of HIF-1α were measured by western blot. Results The mRNA expressions of HIF-1α in As-treated cells were increased significantly at the concentration of 10 μmol / L Na As O2( P〈0. 05); while the antioxidants( NAC and VC) could decrease the protein expression of HIF-1α. Conclusion The results suggest that antioxidants may decrease the risen expression of HIF-1α induced by As.
