摘要
以多花刺槐花粉为实验材料,采用体外培养法,进行了不同保存方式和保存时间处理的花粉离体萌发试验。结果得出,多花刺槐花粉培养2h后开始萌发,培养8h后花粉萌发率达最大值,培养14h后花粉管伸长量达到最大,培养24h后仍有少量花粉萌发,但其活力不足,花粉管伸长较短。随着花粉保存时间延长,萌发率降低;保存4d时萌发率为15%-20%左右,保存6d至8d时萌发率降至9%-10%左右。常温与低温保存相比,保存前期、时间较短时,常温保存的花粉萌发要早,但最终萌发率要低;保存后期,常温保存的萌发优势几乎没有,低温保存在萌发数量与萌发时间上都有明显优势。低温更适宜多花刺槐的花粉保存。
We have do some research about the pollen vitality and the preservation time and ways of Robinia neo-mexicana var.luxurians.Many different in vitro pollen-germination experiments were carried on pollen of Robinia neo-mexicanavar.luxurians.In conclusion,the pollen of Robinia neo-mexicanavar.luxurians began to germinate after two hours'culture and fully germination happened after eight hours'culture then after fourteen hours'culture the pollen tube reached to maximum quantity.And then there maybe also some pollen begin to germinate,but the new pollen tube can't elongate fully,their vitality was too low.As the in vitro culture time increasing,the pollen to germinate lessen,after four days'culture the pollen germination rate was 15%-20%,but when we do the culture after six or eight days,the pollen germination rate was only 9%-10%.Compared to normal temperature and low temperature preservation,at the earlier stage the normal temperature preservation germinated earlier and elongated earlier and at the later stage the low temperature preservation show better vitality.Low temperature maybe better for preservation of pollen of Robinia neo-mexicanavar.luxurians,but we should try to use new pollen in our artificial pollination or hybridization,because while pollen had been preserved its vitality will reduce certainly.
