摘要
目的:观察移动电话辐射对小鼠耳蜗核5-羟色胺受体1B(5-HTR 1B)、5-羟色胺受体2C(5-HTR 2C)基因表达的影响,进而探讨针刺治疗耳鸣的可能机制。方法:30只昆明小鼠随机分为2组,6只作为空白对照组,其余24只接受处于上网及通话状态的移动电话辐射复制耳鸣模型。造模结束后,6只小鼠为模型组,其余18只随机分为模型对照组、翳风穴组(电针双侧"翳风"穴)、肾俞穴组(电针双侧"肾俞"穴),翳风穴组、肾俞穴组每次治疗20min,每日1次,连续7d。RT-PCR法检测各组小鼠耳蜗核5-HTR 1B、5-HTR 2C基因表达情况。结果:与空白对照组比较,模型组及模型对照组小鼠耳蜗核5-HTR 1B明显降低(P<0.05),5-HTR 2 C显著升高(P<0.01);而翳风穴组较模型组耳蜗核5-HTR 1 B明显升高(P<0.01),5-HTR 2C显著降低(P<0.01);与翳风穴组相比,肾俞穴组干预效果较弱(P<0.05)。结论:针刺"翳风"穴可以对小鼠耳蜗核5-HTR 1B、5-HTR 2C基因表达实现良性调整作用,针刺"肾俞"穴效果不明显;针刺调节耳蜗核5-HTR 1B、5-HTR 2C基因表达可能是治疗耳鸣等疾患的作用机制之一。
Objective To observe the effect of electroacupuncture(EA)stimulation of"Yifen"(TE 17),"Shenshu"(BL 23)on the expression of 5-hydroxytryptamine receptor 1B(5-HTR 1B)mRNA and 5-hydroxytryptamine receptor 2C(5-HTR2C)mRNA in the cochlear nucleus tissue in mice experiencing radiation from mobile phone,so as to explore its mechanisms underlying improvement of tinnitus.Methods Thirty Kunming mice were randomly divided into control group(n=6)and modeling group(n=24).The tinnitus model was established by giving the mice with mobile phone-radiation for 1hin the morning and 1h in the afternoon,continuously for 40 days.EA stimulation was applied to"Yifeng"(TE 17)group(n=6)and"Shenshu"(BL 23)group(n=6)for 20 min,once a day for 7days.The expression of 5-THR 1B/2CmRNA in the cochlear nucleus was assayed by fluorescence quantitative polymerase chain reaction(real time-PCR).Results The expression level of 5-HTR 1Bwas significantly lower in the model group than in the control group(P〈0.05),while that of 5-HTR 2C mRNA significantly increased(P〈0.01).TE 17 group received a significant acupoint intervention effect(P〈0.01).Compared with TE 17 group,BL 23 group received a weaker effect(P〈0.05).Conclusion EA of TE 17 can up-regulate expression level of 5-HTR 1Band downregulate expression level of 5-HTR 2Cin the cochlear nucleus in mice experiencing mobile-phone radiation.
