活血除痹汤对硬皮病模型小鼠皮损组织活性氧的影响 被引量:2

Effect of Huoxue Chubi Decoction( 活血除痹汤) on the Active Oxygen of Lesion Tissue of Scleroderma Model Mice

摘要 目的研究活血除痹汤治疗硬皮病的可能作用机制。方法 30只小鼠分为空白组、模型组和中药高、中、低剂量组,每组6只。除空白组外其余各组小鼠皮内注射盐酸博来霉素400μg连续3周建立硬皮病小鼠模型。第4-7周,空白组、模型组予等量蒸馏水灌胃,中药高、中、低剂量组分别给予活血除痹汤44.8、22.4、11.2g/(kg·d)灌胃,每日1次。观察各组小鼠皮肤、肺组织病理学改变,比较皮肤厚度、胶原指数及皮损组织活性氧(ROS)水平。结果模型组小鼠真皮明显增厚,胶原纤维明显增粗,伴有炎性细胞浸润;肺组织出现肺泡间隔增厚伴大量单核细胞浸润,间隙有成纤维细胞增生。与模型组小鼠相比,中药各剂量组真皮胶原纤维增粗膨大程度及肺泡间隔增厚程度均有所减轻。模型组皮肤厚度、胶原指数、皮损ROS含量较空白组明显增加(P〈0.01);与模型组比较,中药高、中、低剂量组皮肤厚度、胶原指数均明显降低,中药高、中剂量组皮损ROS含量亦降低(P〈0.05或P〈0.01)。结论活血除痹汤可明显降低硬皮病模型小鼠皮肤厚度、胶原指数,其机制可能与降低皮损中ROS水平有关。 Objective To study the possible mechanism of Huoxue Chubi Decoction in the treatment of scleroderma.Methods Thirty mice were divided into blank group,model group and traditional Chinese medicine group with high,medium and low dose,6 rats in each group. Except the blank group,the mice were intradermal injected with bleomycin hydrochloride 400 μg for 3 weeks to establish scleroderma mouse model. From the forth week till the seventh week,the rats in blank group and the model group were gavaged with the same amount of distilled water,the rats in traditional Chinese medicine group with high,medium and low dose were unitedly gavaged with Huoxue Chubi Decoction44. 8,22. 4,11. 2 g /( kg·d),once a day. Pathological changes of skin and lung of mice in each group would be observed,skin thickness,collagen index and levels of tissue reactive oxygen species( ROS) would be compared.Results In the model group,the dermis was significantly thickened,and the collagen fibers were increased obviously,and the infiltration of inflammatory cells was accompanied; In the lung tissue,the alveolar septal thickening accompanied with a large amount of monocyte infiltration,and there is fibroblast proliferation. in the gap between the thickening alveolar septal and monocytes. Compared with the model group,the degree of thickening of dermal collagen fibers and alveolar septal were reduced in traditional Chinese medicine group with different dosage. Skin thickness,collagen index and lesion ROS content in the model group were significantly increased than those in the model group( P〈0. 01); Compared with the model group,the skin thickness and collagen index were significantly lower in the high,medium and low dose groups of traditional Chinese medicine,the lesion ROS content of the traditional Chinese medicine group was higher and the middle dose was lower( P〈0. 05 or P〈0. 01). Conclusion Huoxue Chubi Decoction can significantly reduce the skin thickness and collagen index in scleroderma mouse model,the mechanism may be related to decreasing the level of ROS in the lesions.
