左、右归丸对大鼠骨髓间充质干细胞成骨及成脂诱导的研究

Study of Zuogui Pill and Yougui Pill on Osteogenesis and Fat Induced in the Rat Bone Marrow Mesenchymal Stem Cell

目的:研究体外左、右归丸诱导大鼠骨髓充质干细胞(bone marrow stromal stem cells,BMSCs)成骨及成脂的影响。方法:以左、右归丸水煎液和阳性对照药补佳乐、蒸馏水分别给予SD大鼠灌胃取血制备含药血清,分别加成骨和成脂诱导剂干预BMSCs,成骨诱导后采用改良钙钴法检测碱性磷酸酶(Alkaline phosphatase,ALP)表达,成脂诱导第10天油红O染色。结果:左、右归丸组与空白组比较,可显著提高细胞中ALP活性,促进钙化结节形成;左、右归丸可减少脂滴形成,降低细胞中甘油三酯含量。结论:左、右归丸对BMSCs分化的调节具有双向性,既促进BMSCs成骨分化同时又抑制BMSCs成脂分化。

Objective： To investigate the effect of Zuo Gui Wan and You Gui Wan induced on osteogenesis and adipogenic of rats bone marrow stromal stem cells（ BMSCs）. Method： Prepare the medicated serum containing Zuo Gui Wan、You Gui Wan、positive control drug（ Estradiol Valerate） to simultaneously intervene the BMSCs. The ALP test was measured by modified calicium-combat staining method after osteogenic induction. After 10 days of adipogenic induction,oil red O staining of lipid drops was used to detect the adipogenic ability of BMSCs. Results： Zuoguiwan,You Gui Wan,significantly enhanced the ALP activity of the BMSCs and the number of calcified nodules. Zuoguiwan,You Gui Wan significantly reduced the formation of triglyceride levels. Conclusion： Zuoguiwan,You Gui Wan have biphasic regulation of BMSCs,which can promote BMSCs osteogenesis and restrain BMSCs adipogenic at the same time.
