扶正祛邪方抗Con A模型小鼠肝损伤的凋亡机制 被引量:2

摘要 目的:探讨扶正祛邪方对刀豆蛋白A(Con A)诱导小鼠急性肝损伤时肝细胞凋亡的影响及作用机制。方法:清洁级Balb/c雄性小鼠60只,体质量23~25 g,按随机数字表法分为6组每组各10只。连续7 d,每天1次,扶正祛邪方组分别按低(50.3 g/kg)、中(67.0 g/kg)、高(83.8 g/kg)剂量给予小鼠灌胃扶正祛邪方溶液;阳性对照组灌服双环醇溶液(62.5 mg/kg);空白组和模型组灌服等体积生理盐水。于第7天末次给药2 h后,除空白组外其余5组采用Con A(20 mg/kg)尾静脉注射复制肝损伤动物模型,Con A注射6 h后处死动物观测以下指标:小鼠一般状况、肝功能、免疫组织化学观测肝细胞凋亡指数。结果:一般状况模型组肝、脾明显瘀血肿大,肝、脾指数显著增高,高、中剂量FZ可明显减轻小鼠脏器指数及瘀血程度;高、中剂量FZ和双环醇均能显著降低模型小鼠的肝细胞凋亡指数;各给药组均能显著降低肝组织caspase-3表达水平,以高剂量FZ最优。结论:扶正祛邪方可通过减少caspase-3表达而起到抑制Con A模型小鼠肝细胞凋亡的作用,并具有较好的抗Con A模型肝损伤的效果。 Objective: To explore the influence and mechanism of Fuzheng Quxie Prescription on the liver cell apoptosis in mice with Concanavalin A( Con A) induced acute liver injury. Methods: 60 Balb / c male mice weighing 23 to 25 g were randomly divided into 6 groups,with 10 in each group. The solution of Fuzheng Quxie Prescription was given to the mice of high dose group( 83. 8 g / kg),medium dose group( 67. 0 g / kg) and low dose group( 50. 3 g / kg) respectively through intragastric administration daily and continuously for 7 days. The positive control group was given bicyclol solution( 62. 5mg / kg),and the blank group and model group were given equal volume of normal saline. Con A( 20 mg / kg) was given to mice in all groups except the blank control via caudal vein injection 2 hours after the final administration on the 7th day.Six hours after Con A injection,mice were sacrificed to observe the general condition,liver function,the liver cell apoptosis index by immunohistochemistry. Results: 1. General condition: obvious congestion and tumescence of liver and spleen were found in the model group,and the index of liver and spleen markedly increased; the organ index and congestion in the high dose and medium dose groups markedly decreased; The liver cell apoptosis indexes in the high dose and medium dose groups and bicyclol solution group markedly decreased; The expression of caspase-3 declined in each medicated group but most obviously in the high dose group. Conclusion: 1. Fuzheng Quxie Prescription can prohibit the liver cell apoptosis in Con A induced mice by reducing the expression of caspase-3. 2. Fuzheng Quxie Prescription has good anti-injury effect on the mice with Con A induced liver injury.
