摘要
目的:研究针刺捻转补泻手法对SHR(spontaneous hypertensive rats,自发性高血压大鼠)肾脏中TGF-β1(transforming growth factor-β1,转化生长因子-β1)表达的影响及其与肾损害的关系,探讨其保护作用机制。方法:60只12周龄的雄性SHR大鼠被随机分成5组,分别为捻转补法组(A)、捻转泻法组(B)、平补平泻组(C)、针刺留针组(D)、模型组(E),均为高血压组,每组12只;12只12周龄雄性WKY大鼠作为空白对照组。针刺处理28天后,分别取各组大鼠肾脏,观察肾脏病理学改变;应用免疫组化法检测TGF-β1在各组肾脏中的表达。结果:TGF-β1在空白组肾小管的表达无或极微量;在模型组的肾小球有少量表达,但在肾小管的表达显著,且随高血压病程的进展,TGF-β1的表达显著性增加(P<0.01):B组与E组比较有明显差异(P<0.05),A、C、D组之间比较无明显差异(均P>0.05)。结论:捻转泻法是积极有效的降压治疗方法,会减少高压血流对血管内皮和平滑肌细胞的机械性损伤,改善血管内皮功能,减少TGF-β1的分泌,从而降低高血压肾损害的发生率。
Objective: To observe the different effects of reinforcing and reducing methods by twirling and rotating the needle on expressions of TGF-β1in spontaneously hypertensive rats and the relationship with renal damage and to investigate the protective mechanism on kidney damages. Methods: Sixty male SHR rats were randomly divided into 5 groups,12 rats in each group: acupuncture with reinforcing method group(group A),acupuncture with reducing method group(group B),even reinforcing-reducing method group(group C),acupuncture group(group D) and model group(group E). 12 weeks old male WKY rats were as blank control group(group F). After 28 days,taking each kidney of rats,we observed the kidneys pathology change and detected TGF-β1 expression in each group by using immunohistochemical method. Results: TGF-β1 expression in renal tubules of blank group was none or very little. In glomerule,there was a small expression of model group,but the expression of renal tubule was significant and with the progress of the hypertension,the expression of TGF-β1 significantly increased(P〈 0. 01). There was a very significant difference among the contents of TGF-β1 in renal tissue of group B and E(P〈 0. 05). On the contrary,significant differences did not exist among group A,C and D(all P〉 0. 05). Conclusion: Acupuncture with twirling reducing method positive and effective treatment,which can reduce blood flow to reduce mechanical damage of vascular endothelial cells and smooth muscle and improve endothelial function and reduce the secretion of TGF-β1 so as to reduce the incidence of hypertensive renal damage.
