摘要
目的探讨血管内膜障碍对慢性移植物血管病的影响。方法实验分为3组:同系对照组,供、受者均为Lewis大鼠,无处理;异系移植组,Lewis大鼠接受Brown-Norway大鼠腹主动脉移植,分为4周组和8周组。分别于移植后28和56 d取移植动脉,进行组织形态学观察、测量内膜和中膜厚度,免疫组化检测血管内iNOS和PDE-5蛋白表达。结果同系组移植动脉形态正常;异系移植组动脉呈移植物血管病表现,4周开始血管内膜出现显著增厚,随着时间延长,内膜增厚加重(P<0.05)。免疫组化结果显示,与同系移植实验组相比,iNOS表达减弱和PDE-5表达增强。结论抑制NOS信号通路,加重内皮功能障碍,促进移植物血管病变的发展。
Objective The purpose of the study was to study the effects of endothelial dysfunction on chronic allograft vasculopathy. Methods Rat abdominal aorta transplantation was performed on male Lewis or brown- norwai rats as donors and male male Lewis rats as recipients. The recipients were divided into 3 groups: Group A( Lewis- Lewis) that was no treatment; Group B( BN- Lewis) that was no treatment for 4 weeks; Group C( BN- Lewis) that that was no treatment for 8 weeks. Rats were humanely killed at 28 days and 56 days after surgery. The grafted aortas were analyzed by histology and immunohistochemistry. Results The aortas intimal of group C were significantly bodiness compared with group B(P〈0. 05). iNOS protein expression in the group C was significantly lower than those in group B. PDE5 protein expression in the group B and group C were significantly higher than those in the group A. Conclusion Endothelial dysfunction has negative effects of chronic allograft vasculopathy.
