乙酸异戊酯高产菌株的选育及发酵条件优化 被引量:5

摘要 以水果为筛选材料,经过分离纯化、定性试验,从中筛选出1株遗传性状稳定的主要产乙酸异戊酯的酵母菌株Loq-c,经过26SrDNA鉴定,确定其为克鲁维毕赤酵母(Pichia kluyveri)。经过单因素试验及正交试验,菌株Loq-c发酵产酯的最佳条件为:葡萄糖10%,酵母粉4%,KH2PO40.1%,MgSO40.05%,pH 6.0,26℃,150r/min,发酵培养2天,乙酸异戊酯产量最高达到381mg/L。 After purification and qualitative test,1 yeast strain Loq-c yielded of isoamyl acetate is isolated from fruits.According to 26S rDNA gene sequence analysis,strain Loq-c is identified as Pichia kluyveri.After single factor experiment and orthogonal test,the optimum conditions for fermentation and ester-producing of strain Loq-c are:glucose of 10%,yeast extract of 4%,KH2 PO4 of 0.1%,MgSO4 of 0.05%,pH 6.0,26 ℃,150r/min for 2 days,the yield of isoamyl acetate is up to 381 mg/L.
