基于分子进化方法研究HPV与子宫颈癌相互关系 被引量:1

Research on the relationship between HPV and cervical cancer based on molecular evolution methods

摘要 目的阐述不同类型HPV与子宫颈癌的关系,对潜在高危型HPV做出预测,为临床宫颈癌的早期预防和诊断提供理论依据。方法从NCBI等数据库下载已发表的131种不同基因型的HPV序列,构建出HPV基因序列数据集。应用MEGA软件的neighbor—joining法对HPV全基因组及各基因片段数据集进行了分子进化分析,构建出系统亲缘进化树。结果目前全球HPV根据全基因组分子进化分析可以分为五大类,与宫颈癌密切相关的高危型HPV高度聚集在一起。黏膜亲和型HPV各基因片段分析显示,以E6、E7基因区域构建的进化树更接近于全基因组进化树的分布。预测了HPV26、34、53、66、67、97对宫颈癌的发生有潜在危险性。结论本研究构建了不同亚型HPV的数据集,根据分子进化方法分析得到的HPV亲缘性与子宫颈癌的关系与临床相一致,为宫颈癌相关HPV的早期预防和诊断提供理论依据。 Objective We aimed to analyze the relationship among all types of HPV and cervical cancer and predict potential high-risk HPV based bioinformatics methods. Provide theoretical basis for clinical prevention and early diagnosis for cervical cancer. Methods A total of 131 types HPV gene sequences from GenBank were downloaded. HPV phylogenetic tree were constructed use the MEGA software. Results Current global HPV can be divided into five categories according to complete gene molecular evolution analysis, high-risk types HPV are height together. Further to the mucous membrane affinity types HPV gene analysis shows that the evolution of the E6, E7 gene closer to the distribution of the whole genome evolution tree. We predict the HPV 26,34,53,66,67,97 have potential dangers with cervical cancer. Conclusion Building the HPV data based on the analysis of molecular evolution method of HPV affinity relationship with cervical cancer. Provide theoretical foundations for HPV clinical prevention and treatment of cervical cancer.
