摘要
本研究通过免疫组化及活细胞计数法研究了苯丙氨酸及其代谢物对 P19细胞神经分化的影响。结果显示 :在 P19细胞神经分化诱导期加入苯丙氨酸及其代谢物可降低 P19神经元存活率 ,导致 P19神经元肿胀、崩解 ,神经突起纤细且分支少 ,神经纤维丝蛋白的染色性降低 ,同时发现 P19神经元中多极神经元 /单双极神经元的比例下降。本研究表明 ,苯丙氨酸及其代谢物可影响 P19细胞的神经诱导和分化 。
The effect of phenylalanine (PHE) and its metabolites on the neuronal differentiation of P19 embrional carcinomal cell (P19 cell)was studied by immunohistochemistry and counting of survival P19 neurons. The results indicated that P19 neurons exposed to PHE and its metabolites phenylacetic acid (PAA), phenyllactic acid (PLA) from induction phase of P19 neuronal differentiation showed various changes such as cell swelling, collapsing, slender neurofibrils, decrease in the number of neurofibrils, weaker NF staining and so on. The viability of P19 neurons was remarkably decreased. The ratio of multipolar neurons to bipolar/unipolar neurons was also lower than that of the control group. Our data showed a primary effect of PHE and its metabolites on P19 cell neuronal differentiation, and suggest PHE or its metabolites might be involved in the central nervous system dysfunction associated with maternal PKU.
