摘要
为探讨成人海马内 Nestin免疫阳性细胞的分布 ,本实验采用 331B、10 C2、Rat40 1、NSE、GFAP、Vimentin抗体对成人海马了进行免疫组织化学研究。结果显示 ,海马内 Nestin免疫阳性细胞可分为三类 :一类细胞位于门区及齿状回颗粒下层 ,胞体为卵圆形 ,无突起 ,胞浆深染 ,此类细胞不与 NSE呈交叉反应 ;第二类细胞也位于门区及齿状回颗粒下层 ,胞体较小发出少量放射状突起 ,与 GFAP及 Vimentin抗体无交叉反应 ,在海马裂及海马伞中也存在少量的此类神经细胞 ;第三类为 Nestin免疫阳性细胞 ,体积较大 ,形态与星形胶质细胞相似 ,并分别位于齿状回分子层及颗粒层 ,其突起常常跨越齿状回颗粒细胞层。Nestin免疫阳性神经细胞不被 GF AP抗体标记。成人海马内未见 Vimentin免疫阳性细胞。结论
To investigate the distribution and chemical character of nestin immunoreactive cell in the hippocampus, we explored systematically the distribution of nestin immunoreactive cells throughout the adult human hippocampus by 331B, 10C2, Rat401 antibody. Furthermore, we investigated the chemical identity of these nestin immunoreactive cells by using double-labeling immunocytochemistry with NSE, GFAP and vimentin antibody. The nestin immunoreactive cells in hippocampus could be divided into three groups. The first group cells were located in the hilus and subgranular layer of the dentate gyrus. The cells were oval-shaped with dark stain of cytoplasma and had no processes. The second group cells were located in hippocampal fissure, fimbria, and subgranular layer of the dentate gyrus. The cells were very small and contained a few radial processes. The third group cells were large and shared similar morphology with that of astrocyte glia. They were located in the stratum molecular of the dentate gyrus and their processes always crossed the granular layer of dentate gyrus. All the nestin positive cells could not be labeled by NSE, GFAP or Vimentin antibody. These results suggest that adult human hippocampus contain neuroprogenitor cells or radial glia-like nesin immunoreactive cells. (Figures 1~6 on plate 33)
