摘要
目的阐明面部穴位埋线通过调节变应性鼻炎动物模型鼻腔黏膜最轻持续性炎症状态(minimal persistent inflammation,MPI)进而延长变应性鼻炎的缓解期,降低其复发率的机理。方法采用卵清蛋白致敏SD大鼠的方法制作变应性鼻炎MPI动物模型,分为治疗组A(穴位埋线干预的变应性鼻炎MPI组)和对照组B(变应性鼻炎MPI组),另设空白组C。A组迎香穴(LI20)埋线治疗。观察内容:1)各组大鼠变应性鼻炎相关的行为学评分变化;2)A组埋线后2周,取大鼠鼻黏膜,应用组织病理学和免疫组织化学方法观察鼻黏膜嗜酸性粒细胞(eosinophil,EOS)及细胞间黏附分子-1(intercellular adhension molecule-1,ICAM-1)的改变。结果 A组大鼠迎香穴埋线后,三组动物变应性鼻炎相关的行为学评分差异无统计学意义。苏木素-伊红染色(hematoxylin-eosin staining,HE染色)显示穴位埋线治疗能够改善鼻黏膜MPI状态下EOS的表达,并接近于正常鼻黏膜。免疫组织化学检测,A、B、C三组大鼠鼻黏膜的ICAM-1积分光密度值经两两比较,差异均具有统计学意义。结论面部穴位埋线通过改善以EOS及ICAM-1为标志的变应性鼻炎鼻黏膜最轻持续性炎症状态,并由此可能延长了变应性鼻炎的缓解期,降低其复发率。
Objective To determine the desensitization effect of catgut implantation at acupoint on experimental minimal persistent inflammation( MPI) models in allergic rhinitis( AR) rats. Methods Thirty ovalbumin( OVA)-sensitized rats with minimal persistent inflammation of allergic rhinitis were randomly assigned into the study group( group A) or the control one( group B).And another 15 rats were used as no-sensitized and no-treatment group( group C). When rats in group A had been treated with catgut implantation at Yingxiang( LI20) for 2 weeks,the scores of allergic symptoms were assessed and the changes of eosinophils( EOS) and intercellular adhension molecule-1( ICAM-1) in the nasal mucosa were studied by pathological and immunohistochemical methods. Results Scores of symptoms of allergic rhinitis showed no statistical significance between the three groups. Immunohistochemical studies revealed a marked decrease of ICAM-1 in the nasal mucosa between group A and group B. Hematoxylin-eosin( HE) staining demonstrated that the number of eosinophils decreased in the nasal mucosa of group A. Conclusion The results suggest that catgut implantation at acupoint can regulate the MPI of allergen-induced nasal inflammatory cascade in rats.This treatment is helpful in prolonging the remission stage of AR and reducing the recurrence of this disease.
