转化生长因子β1对种植体表面成骨细胞中骨钙素基因表达研究 被引量:4

EFFECTS OF TGF-Β1 ON THE LEVEL OF OSTEPOCALCIN MRNA IN OSTEOBLASTS ON IMPLANT SURFACE

摘要 目的通过观测不同浓度转化生长因子β1(transforming growth factorsβ1,TGF-β1)对种植体表面成骨细胞中骨钙素(osteocalcin,OC)表达的影响,以探讨其对种植体表面成骨细胞分化的作用。方法专用钛片模拟种植体植入后的体内环境,体外培养成骨细胞接种于钛片表面,分为实验组和对照组,实验组分别加入0.5ng/ml、10ng/ml的TGF-β1,收集第1d、3d、7d培养后的细胞,对照组不加TGF-β1。通过RT-PCR检测不同培养时间成骨细胞中OCmRNA的表达情况。所有数据采用SPSS 16.0软件作统计学处理,然后比较分析各组间差异程度,P<0.05认为差异有显著性意义。结果浓度为0.5ng/ml外源性TGF-β1加入到接种于钛片表面的成骨细胞中后,显著增加了成骨细胞中骨钙素的表达(P<0.05),而浓度为10ng/ml的外源性TGF-β1可降低成骨细胞中骨钙素的表达(P<0.05)。结论外源性TGF-β1对接种在模拟体内种植体钛片表面成骨细胞中OC表达的作用与其浓度相关。 Objective By observing different concentration of transforming growth factor beta 1(transforming growth factors beta 1,TGF-β1)of the implant surface osteoblast in osteocalcin(osteocalcin,OC)expression and the effect of in order to investigate its effect on osteoblast differentiation on the surface of the implant.Methods Osteoblast were divided into two groups:the control group and the induced group.The cells were collected on Days 1,3,7after treatment.The expression of osteocalcin(BSP)mRNA were detected by reverse transcription-polymerase chain reaction(RT-PCR).All the data were analyzed by SPSS16.0software.Results The extraneous(0.5ng/ml)can increase the gene expressions of osteocalcin in osteoblasts on implant surfaces(P 0.05),but reduced when treatment with 10ng/ml of TGF-β1(P 0.05).Conclusion Exogenous TGF-β1docking in simulated body titanium implant surface OC expression in osteoblast function is related to its concentration.
