摘要 转化生长因子β1(TGF-β1)在肝纤维化的发生、发展中起重要作用,它作为TGF-β1/Smad信号通路的起始因子激活下游信号,使处于非活性状态下的肝星状细胞分化为肌成纤维细胞,促进并维持了肝纤维化的发生。泛素化是体内蛋白质翻译后修饰并降解的重要途径之一,泛素-蛋白酶体途径中的泛素连接酶Smurf2通过多种途径抑制TGF-β1/Smad通路表达。Smurf2作为一种抗肝纤维化发生的酶受到广泛关注,但机制并未深入系统地被阐述。 Transforming growth factor β1 ( TGF-β1 ) plays an important role in the development of liver fibrosis, as an initiative factor of TGF-β1/Smad pathway, it could activate downstream signals, which makes the inactive hepatic stellate ceils to differentiate into myofibroblasts, promoting and maintaining the state of fibrosis. Ubiquitination is one of the important in vivo ways of translational modifications and protein degrada- tion, and Smurf 2 ubiquitin ligase in ubiquition-proteasome pathway suppresses the expression of TGF-β1/ Smad pathway through a variety of ways. Smurf 2 as an anti-hepatic fibrosis enzyme has attracted wide atten- tion, but the mechanisms have not been systematically elaborated.
