摘要
谷胱甘肽过氧化酶(Glutathione peroxidase,GPx)可以清除过量的氢过氧化物以保持活性氧(ROS)代谢平衡使机体免受损伤,作为一种抗氧化剂具有一定的药用价值。然而天然GPx异源表达困难、酶水解以及半衰期短等缺点限制了其应用,因而设计合成高效GPx抗氧化酶模拟物受到学术界广泛关注。本文从化学修饰法和基因工程法两个方面介绍了GPx人工模拟酶的主要合成方法。通过比较已有GPx人工模拟酶的优缺点及活力值,对其发展趋势做出展望。
Glutathione peroxidase(GPx) can remove the excess hydrogen peroxide to maintain the reactive oxygen metabolic balance to protect the body from injury. It has some medicinal value as an antioxidant. However, the disadvantage such as difficult heterogenous expression, solution instability, short half - lives and proteolytic digestion limited the application of the naturally GPx. Enormous efforts have been made to simulate the highly efficient functions of GPx. This paper introduced two methods including chemical modification and genetic engineering to synthesis GPx artificial enzyme. By comparing the activities of the GPx mimics,we could identify the current developing situation and its future development.
