凋亡相关基因膜转运蛋白166启动子区CpG岛甲基化在新疆汉族和维吾尔族食管鳞状细胞癌中作用 被引量:2

Role of apoptosis-related gene transmembrane protein 166 promoter methylation in Han and Uygur patients with esophageal squamous cell carcinoma in Xinjiang

摘要 目的探讨新疆维吾尔族(维族)和汉族食管鳞状细胞癌(鳞癌)患者凋亡相关基因膜转运蛋白166(transmembrane protein 166,TMEM166)启动子区CpG岛甲基化的差异,及其与食管鳞癌临床病理特征及预后的关系。方法 40例食管鳞癌患者,其中汉族和维族各20例,采用甲基特异性PCR法检测癌组织和癌旁组织TMEM166启动子区CpG岛甲基化情况,分析TMEM166基因甲基化与食管鳞癌临床病理及预后的关系。结果癌组织TMEM166启动子区CpG岛甲基化阳性率(75.0%)明显高于癌旁组织(45.0%)(P<0.05);维族TMEM166启动子区CpG岛甲基化阳性率(70.0%)与汉族(80.0%)比较差异无统计学意义(P>0.05);食管鳞癌Ⅲ期患者TMEM166启动子区CpG岛甲基化阳性率(95.0%)明显高于Ⅱ期患者(51.0%)(P<0.05);食管鳞癌患者TMEM166启动子区CpG岛甲基化阳性率在民族、性别、年龄上差异无统计学意义(P>0.05);TMEM166启动子区CpG岛甲基化阳性者中位生存时间为28个月,阴性者为27个月,差异无统计学意义(P>0.05)。结论食管鳞癌组织TMEM166启动子区CpG岛甲基化水平高于癌旁组织,TMEM166启动子区CpG岛甲基化阳性率可能与食管鳞癌TNM分期有关,与患者年龄、性别、民族和预后无关。 Objective To investigate the ethnic differences of apoptosis-related gene transmembrane protein 166 (TMEM166) promoter CpG island methylation between Uygur and Han patients with esophageal squamous cell cancer in Xinjiang, and its relationship with clinicopathological characteristics and prognosis of esophageal squamous cell cancer. Methods Forty patients with esophageal squamous cell cancer including 20 Uygur and 20 Han patients were detected the methylation of TMEM166 in esophageal squamous cell cancer tissue and the adjacent normal tissue by using PCR technique to analyze the relationship between methylation and prognosis. Results The positive rate of TMEM166 methylation was significantly higher in cancer tissue (75.0 % ) than that in adjacent normal tissue (45.0% ) (P〈0.05), and higher in stage [[I (95.0%) than that in stage ]1 (51.0~~) in patients with esophageal squamous cell cancer (P〈0.05). The positive rate of TMEM166 methylation showed no significant difference between Uygur (70.0%) and Han patients (80.0%) (P〉0. 05), and between different genders and ages (P〉0.05). The mean survival time was 28 months in patients with positive TMEM166 methylation and 27 months in negative TMEM166 methylation, showing no significant difference (P〉0.05). Conclusion The methylation level of TMEM166 is higher in esophageal squamous cell carcinoma tissue than that in the adjacent normal tissue. The positive rate of TMEM166 methylation is correlated with the TNM staging of esophageal squamous cell carcinoma, but not correlated with the age, gender, ethnics or prognosis.
