摘要
研究探讨了脂肪分化相关蛋白(Adipocyte differentiation-related protein,ADFP)基因SNPs与屠体性状的相关性。运用PCR-SSCP技术进行多态性分析,结果表明:在三穗鸭ADFP基因外显子1的248 bp处发现1个单链构象多态,为T→C突变,得到三种基因型(CC、TT、CT),且在此位点的优势等位基因型为TT基因型,T为优势等位基因,在外显子2、外显子3、外显子4、外显子5、外显子7并未发现突变位点。三穗鸭屠体性状显示,公母鸭之间的全净膛重差异极显著(P<0.01);屠体重、腿重、翅膀重有显著差异(0.01<P<0.05)。通过关联性分析,公鸭CC基因型的个体半净膛重显著大于TT和CT基因型个体(P<0.05),母鸭TT基因型个体胸肌重显著高于CC和CT基因型个体(P<0.05);其它屠体性状无显著差异(P>0.05)。
To study the association between single nucleotide polymorphism sites (SNPs)of adipocyte differentiation- related protein (ADFP) gene and slaughter traits in Sansui duck, the polymorphism of those genes was analyzed by PCR-SSCP. Sequencing results showed that there was a T→C mutation in exonl at 248 bp of ADFP gene,and three genetypes (CC,TT, CT). Allele T was identified as the preponderant gene and TT was preponderant genotype at the site. Mutation sites were not found in exon2, exon3, exon4, exon5 and exonT. The carcass traits of Sansui duck indicated that all-eviscerated weight was significantly different (P〈0.01), and carcass weight, leg muscle weight and wings weight were significantly different among different gender ducks (0.01〈P〈0.05). Through correlation analysis,the semi-eviscerated weight of CC genotype individual was significantly higher (P〈0.05) than that of TT, CT genotype in male ducks,chest muscle weight of TT genotype was significantly higher than that of CC and CT genotype. The other carcass traits had no significant difference (P〉0.05).
