摘要
[目的]观察细胞黏附分子CD146在乳腺癌细胞SKBR3中过表达对裸鼠体内肿瘤生成的影响.[方法]采用脂质体转染乳腺癌细胞株SKBR3,经G418筛选获得过表达CD146阳性克隆细胞,将其接种于裸鼠皮下组织,观察并记录肿瘤生长状况;给肿瘤组织行免疫组织化学染色,观察CD146在肿瘤组织中的表达水平.[结果]将SKBR3细胞经脂质体转染与G418筛选后获得的过表达CD146阳性克隆细胞接种于裸鼠皮下组织后可促进肿瘤组织在裸鼠体内的生成;免疫组织化学染色观察结果表明,接种过表达CD146的SKBR3细胞生成的肿瘤组织仍可高表达CD146蛋白分子.[结论]过表达CD146的乳腺癌细胞SKBR3可促进裸鼠体内的肿瘤生成.
OBJECTIVETo investigate the effects of overexpression of cell adhesion molecule CD146 of breast cancer cell line SKBR3 on tumorigenesis in nude mice in vivo.METHODSLipofection of SKBR3 was performed and the transfectant was screened by G418 resistence to obtain positive clone cells that overexpressed CD146.These cells were subcutaneously injected into subcutaneous tissue of the nude mice,and the tumorigenesis of SKBR3 was observed and recorded every day.Expression of CD146 in tumor tissue of nude mice was detected by immunohistochemistry.RESULTS After lipofection and G418 screening,the positive clone cells with CD146 overexpression could promote tumor formation in the nude mice after subcutaneous injection,and tumor tissue from nude mice injected with positive SKBR3 cells still could express high level of CD146 protein.CONCLUSIONBreast cancer cell SKBR3 with CD146overexpression can promote the tumorigenesis in nude mice in vivo.
