Effect of mouse β-defensin-1 on the Hek293 cells

摘要 目的观察经原核表达得到的小鼠β-防御素-1(mBD-1)对Hek293细胞中CCR6-CCL20通路的影响以及对促进Hek293细胞趋化移动的情况。方法用定量PCR方法检测CCR6和CCL20的基因表达情况,同时用Transwell小室研究其对Hek293细胞的定向移动作用。结果 mBD-1可以使CCR6的表达量增加,且mBD-1浓度越高时CCR6表达量越高,差异有统计学意义(P<0.05)。CCL20的表达量不随mBD-1浓度的升高而变化。同时mBD-1可以促进Hek293细胞作定向移动。结论本研究为进一步研究mBD-1对细胞的免疫诱导机制奠定基础。 Objective The effect of mBD-1(conserved in my lab and obtained from BL21) on the CCR6-CCL20 and chemotactic on the Hek293 cells were studied. Methods The expression level of mRNA was detected by q PCR. The Hek293 cell migration was detected by transwell assay. Results The results showed mBD-1 can induce the CCR6 expression and invasion activity in Hek293 cells, and expression of CCR6 statistically significant increased with the increase of m BD-1 concentration. Conclusion This study provided a basic for mBD-1 research.
