血平板辅助检出急性腹泻患者粪便标本中O139霍乱弧菌

Determination of O139 Vibrio cholera in stool samples from patients with acute diarrhea aided by blood plate

摘要 目的研究在使用广谱抗生素治疗后的急性腹泻患者粪便标本中,分离培养霍乱弧菌的方法。方法将使用抗生素治疗后的急性腹泻患者粪便标本在接种碱性蛋白胨水和TCBS平板、庆大霉素平板等进行常规细菌分离培养的同时,接种血平板辅助培养,于37℃培养10 h^12 h后挑取可疑菌落再转种碱性琼脂平板,于37℃继续培养。结果接种血平板37℃培养10 h后形成直径2 mm左右的可疑菌落,再转种碱性琼脂平板37℃继续培养10 h后分离得到O139霍乱菌株,而直接接种碱性蛋白胨水、TCBS平板和庆大霉素平板等常规方法分离培养12 h仍无菌生长。结论利用血平板辅助培养可提高使用广谱抗生素治疗后的急性腹泻患者粪便标本中霍乱弧菌分离培养成功率,减少因抗生素抑菌作用而导致的漏检。 Objective To study the method of separating and culturing Vibrio cholerae O139 from feces of patients with acute diarrhea after antibiotic treatment. Methods The samples of feces from patients with acute diarrhea after antibiotic treatment were inoculated on alkaline peptone water,TCBS plate,gentamicin plate for the regular separating and culturing of bacteria,at the same time which was also inoculated on the blood palate for assistant culturing. And under 37 ℃,10 h- 12 h later,the possible bacterial colony were selected and transfered on alkaline agar plate to continue nurturing. Results 10 h later,the suspicious colonies formed on blood plate with diameter of about 2 mm,and 10 h later,Vibrio cholerae O139 was obtained from alkaline agar plate,but no bacterium grew on alkaline peptone water,TCBS plate,and gentamicin plate after 12 h. Conclusion Aided by blood plate can help improve the success rate of isolation and culture of Vibrio cholerae O139 from feces of patients with acute diarrhea after antibiotic treatment,and reduce mistakes caused by antibiotics antibacterial.
