摘要
目的建立检测血清中十八碳三烯酸(ALA)、二十碳五烯酸(EPA)2种多不饱和脂肪酸的新方法,并用该方法检测危重症患者与正常人血清中ALA、EPA的含量。方法将人血清中的十八碳三烯酸(ALA)、二十碳五烯酸(EPA)经正己烷萃取浓缩,经硫酸-甲醇甲酯化,用气相色谱法定性、定量检测。结果危重患者和正常人血清中的ALA、EPA含量差异均无统计学意义(P>0.05)。检出限(S/N=3):ALA:2.3μg/ml,EPA:1.4μg/ml。ALA,EPA的RSD(n=6)分别为2.32%、3.94%。结论此方法重复性好、敏感度高,可用于Omega-3多不饱和脂肪酸含量的测定。
Objective To establish a new method for determining the contents of two polyunsaturated fatty acids octadecadienoic acid( ALA) and eicosapentaenoic acid( EPA) and to determine the contents of EPA and ALA in serum of critically ill patients and healthy persons using this method. Methods Octadecadienoic acid and eicosapentaenoic acid in human serum were were extracted and concentrated by hexane and methylesterified by H2SO4- CH3 OH,and qualitative and quantitative detection were conducted by gas chromatography. Results There was no significant difference in the content of ALA and EPA in the serum of the patients with severe patients and normal persons( P〈0. 05). The limit of detection( S / N = 3) : ALA: 2. 3 μg / ml,EPA:1. 4 μg / ml. RSD of ALA and EPA( n = 6) were 2. 32%,3. 94%. Conclusion This method has good repeatability and sensitivity,and can be used for the determination of Omega- 3 polyunsaturated fatty acids.
