摘要
以牦牛气管为原料,采用超声波辅助碱处理提取硫酸软骨素,研究了料液比、超声波功率、超声时间对其得率的影响,通过正交试验优化了提取工艺。结果表明,超声波辅助碱提取牦牛气管中硫酸软骨素的工艺条件为:料液比1∶6(g/m L),超声波功率为550 W,超声时间为30 min,在提取温度35℃,p H 12的条件下硫酸软骨素的得率可达18.97%。各因素对硫酸软骨素得率的影响顺序依次为:超声功率>超声时间>料液比。比较了TCA法、Sevage法、胰蛋白酶法的除蛋白效果,其中胰蛋白酶法效果最佳,蛋白质去除率可达85.78%,硫酸软骨素纯度达85.56%。
Use ultrasonic wave to extract chondroitin sulfate fi'om yak trachea, research on the effect of solid-liquid ratio, ultrasonic power and ultrasonic treatment time on chondroitin sulfate the yield. The technological optimum parameters were found as follows: solid-liquid ratio was 1 : 6 (g/mL), ultrasonic power was 550 W, ultrasonic treatment time was 30 min, and extraction temperature was 35 ~C, pH 12, under these conditions, chondroitin sulfate the yield could reach 18.97%. The primary and secondary factors which affected the yield showed as following: ultrasonic power〉ultrasonic treatment time〉 solid-liquid ratio. Three deproteinization methods: trichloroacetic acid method, Sevage method and parenzyme method were compared. It showed that the parenzyme method was the best, its protein removal rate could reach 85.78%, and purity of chondroitin sulfate was 85.5%A.
