虾夷马粪海胆生殖腺多糖形成多聚体现象研究

摘要 在提取海胆生殖腺多糖过程中发现提取产物存在多糖聚合现象。为揭示多糖聚合原因,以Sephadex G-75对多糖进行纯化,将得到的海胆生殖腺多糖SUGP进行高效液相色谱法分析。试验结果表明:SUGP在氯化钠盐溶液存在的情况下在液相色谱图中出现单一峰,为纯度良好多糖;但在去除氯化钠盐离子后,SUGP在液相色谱图中出现两个分子量大小不同的检测峰。分子量检测结果表明SUGP单体的分子量为4 436 Da。PMP柱前衍生化高效液相色谱法分析SUGP单糖组成表明,SUGP单糖组成为甘露糖、葡萄糖和氨基葡萄糖。 Polysaccharide extracted from gonad of sea urchin was found to have aggregation phenomena during the purification processing. To reveal this phenomenon, the polysaccharides were purified with a column of Sephadex G-75. The product of SUGP was analysis by High Performance Liquid Chromatography. The result showed that SUGP produced a single peak in the solution of sodium chloride. However, SUGP produced several peaks with different molecular weight when sodium chloride was removal from the solution. The molecular weight of monomer SUGP was determined to be 4 436 Da. SUGP contained of marmose, glucose and amino glucose analyzed by PMP derivatization method.
