摘要
目的探讨细胞外信号调节激酶/p38丝裂原活化蛋白激酶(ERK/p38MAPK)信号通路在参苓白术散调控脾虚湿困型溃疡性结肠炎大鼠结肠组织水通道蛋白(aquaporin,AQP)3、AQP4表达中的作用。方法将60只Wistar大鼠按照随机数字表均分为正常组、模型组(氯化钠注射液)、参苓白术散组、U0126+参苓白术散组、SB203580+参苓白术散组。除正常组外,脾虚湿困型溃疡性结肠炎大鼠采用2,4,6-三硝基苯磺酸/乙醇灌肠,结合环境与饮食干预复制。14 d后采用蛋白质印迹法检测各组大鼠结肠组织AQP3、AQP4蛋白的表达,实时荧光定量PCR法检测其mRNA表达情况。结果模型组大鼠AQP3、AQP4蛋白表达及其mRNA的表达水平明显低于正常组(P<0.05),参苓白术散组大鼠结肠组织AQP3、AQP4蛋白及mRNA表达较模型组明显升高,组间比较有显著性差异(P<0.05),SB203580+参苓白术散组及U0126+参苓白术散组大鼠结肠组织AQP3、AQP4蛋白及mRNA表达较模型组有较小幅度升高,与正常组和参苓白术散组相比均有显著性差异(P<0.05)。结论参苓白术散可显著改善大鼠结肠组织AQP3、AQP4蛋白及mRNA表达,ERK/p38 MAPK信号通路参与了参苓白术散对脾虚湿困型溃疡性结肠炎大鼠结肠组织AQP3、AQP4表达的调节作用。
AIM To investigate the effects of Shenlin Baizhu Powder on the expressions of AQP3 and AQP4 in ulcerative colitis rats with syndrome of dampness retention and spleen deficiency via ERK / p38 MAPK signal pathway. METHODS Sixty rats were randomly divided into normal group,model group( receiving sodium chloride injection),Shenlin Baizhu Powder group,U0126 + Shenlin Baizhu Powder group and SB203580 + Shenlin Baizhu Powder group. The model rats were induced by environment and diet intervention combined with the perfusion of TNBS and ethanol except for the normal group. In the 14^th day,the protein expression changes of AQP3 and AQP4 in rats were detected by Western blot. Their mRNA expression changes were detected by Fluorescence quantitative PCR. RESULTS The proteins and their mRNA expressions of AQP3 and AQP4 in the model rats were lower than those of the normal group( P 0. 05). The expressions of these proteins and mRNA in Shenlin Baizhu Powder group increased significantly( P 0. 05). SB203580 + Shenlin Baizu Powder group and U0126 + Shenlin Baizu Powder group increased in a smaller amplitude as compared with Shenlin Baizhu Powder group and normal group with significant difference( P 0. 05). CONCLUSION Shenlin Baizhu Powder can significantly improve the expressions of AQP3 and AQP4 in colon tissues. ERK / p38 MAPK signal pathway is involved in the regulation of Shenlin Baizhu Powder on the expressions of AQP3 and AQP4.
