摘要
目的探讨抑癌基因p53及分化抑制因子-1(Id-1)在人类宫颈组织中的表达及其临床意义。方法采用实时荧光定量PCR及免疫组化(IHCA)SP二步法方法,分别检测64例子宫颈鳞状上皮癌(SCC)、34例宫颈上皮内瘤变(CIN)组织及30例正常宫颈组织p53和Id-1 mRNA及蛋白表达水平,分析两者在宫颈癌组织中表达的相关性。结果 p53 mRNA在SCC组织中表达量是正常宫颈组织的2.638倍,是CIN组织中的1.611倍,在CIN组织中的表达量是正常宫颈组织中的1.605倍,差异有统计学意义(P<0.05);p53蛋白在正常宫颈组、CIN组及SCC组阳性表达率分别为0%、52.9%、71.9%,差异有统计学意义(P<0.05)。Id-1 mRNA在SCC组织中的表达量是正常宫颈组织中的3.22倍、是CIN组织中的1.137倍,在CIN组织中的表达量是正常宫颈组织中的1.862倍,差异有统计学意义(P<0.05);Id-1蛋白在正常宫颈组、CIN组及SCC组阳性表达率分别为0%、50.0%、73.4%,差异有统计学意义(P<0.05)。对p53及Id-1的mRNA表达量做Spearman等级相关分析,差异有统计学意义(P<0.05);对p53及Id-1的蛋白表达量差异有统计学意义(P<0.05)。结论 p53、Id-1与宫颈癌的发生发展密切相关,且两者表达具有相关性。
Objective To investigate the expression of tumor suppressor gene p53 and oncogene Id-1 in the tissues and clinical significance of cervical cancer by human beings. Methods 128 patients aged between 33- 74 years old were divided to 30 normal cervical tissues,34 cervical intraepithelial neoplasia tissues and 64 cervical cancer by pathological examination. The real- time fluorescence quantitative PCR and immunohistochemical staining(SP) were used to detect the expression of p53 and Id- 1 in cases of tissues respectively,analyzed their relationship on expression of cervical cancer. Results The PCR showed the expression of p53 mRNA in cervical squamous cell was 6. 503 times that in normal cervical tissues,and it was 2. 201 times that in CIN tissues,in CIN tissues was 2. 955 times that in normal cervical tissues,with a statistically significant difference(P〈0. 05); the IHCA showed the positive rate for p53 expression in the control group,the CIN group and the SCC group was respectively 0%,52. 9% and 71. 87%(P〈0. 05). The expression of Id- 1 mRNA in SCC tissues was 2. 734 times that in normal cervical tissues,and it was 1. 137 times that in CIN tissues,in CIN tissues was 2. 144 times that in normal one,with a statistically significant difference(P〈0. 05); The IHCA showed the positive rate for Id- 1 expression in the control group,the CIN group and the SCC group was respectively 0%,50. 00% and 73. 44%(P〈0. 05). Id- 1 expression was positively correlated with p53 expression in the process of carcinogenesis of cervical squamous cell carcinoma. Conclusion The impression of Id- 1 and p53 were enhanced from normal tissues to cancer tissues no matter in PCR or IHCA method,remind us if Id- 1 and p53 play important role in the occurrence and development of cervical cancer.
