塞来昔布联合放射治疗对异位裸鼠人鼻咽癌模型中肿瘤细胞增殖与凋亡的影响

摘要 目的:探讨塞来昔布联合放射治疗对异位裸鼠人鼻咽癌模型中肿瘤细胞增殖与凋亡的影响。方法:建立裸鼠人鼻咽癌皮下移植模型,成瘤后40只裸鼠随机分成4组,即对照组、塞来昔布组、放射治疗组、联合组(塞来昔布与放射治疗),观察肿瘤大小,计算抑瘤率;采用RT-PCR方法考察肿瘤中COX-2的mRNA表达,并用免疫组化法以Ki-67为指标测肿瘤细胞增殖水平,末端脱氧核苷转移酶标记法(TUNEL)测凋亡细胞。结果:对照组、塞来昔布组、放射治疗组、联合组瘤体体积分别为(1880±358)、(1574±265)、(1258±262)、和(881±197)mm3;塞来昔布组、放射治疗组、联合组抑瘤率分别为16%、33%和53%。四组中Ki-67阳性率分别为44%、29%、22%和11%;肿瘤细胞凋亡指数(AI)分别为9%、22%、30%和38%;塞来昔布组、联合组肿瘤组织中COX-2 mRNA表达显著低于对照组和放射治疗组。结论:塞来昔布可显著抑制裸鼠人鼻咽癌模型中肿瘤细胞COX-2 mRNA表达,抑制肿瘤细胞Ki-67表达,诱导肿瘤细胞凋亡,与放射治疗联合应用可提高抗肿瘤效果。 AIM: To investigate the effect of celecoxib combined with radiotherapy on proliferation and apoptosis of nasopharyngeal carcinoma in nude mice model. METHODS: CNE-2 nasopharyngeal tumor cells were implanted in 40 nude mice and the mice were divided randomly into 4 groups:the control group,the celecoxib group,the radiotherapy group and the celecoxib combined with radiotherapy group. RESULTS: Tumor volume was( 1880 ± 358),( 1574 ± 265),( 1258 ± 262) and( 881 ± 197) mm3 in the control,celecoxib,radiotherapy and combine groups,respectively. The rate of inhibition for tumor volume was 16% in the celecoxib group,33% in the radiotherapy group and53% in the combined group. Immunohistochemical assay for tumor proliferation( Ki-67) showed that the tumor cell Ki-67 positive rate was respectively44%,29%,22% and 11% and apoptosis index was 9%,22%,30% and 38%,respectively. The expression of COX-2 mRNA by RT-PCR in celecoxib group and combined group was highly suppressed compared with that of the control group and radiotherapy group. CONCLUSION: Celecoxib can suppress the expression of COX-2 mRNA of tumors implanted in nude mice,and reduce proliferation and induce apoptosis of tumor cells; by combined with radiotherapy,it can improve its anti-tumor activity.
