摘要 目的:研究MTNR1B rs10830963基因多态性对瑞格列奈治疗2型糖尿病(T2DM)患者疗效的影响。方法:选取新诊断的T2DM患者90例,口服瑞格列奈1 mg,一日3次,连续8周,服药前和服药第8周末收集静脉血标本,测定治疗前后人体参数及糖脂代谢相关指标。采用高分辨溶解曲线方法对患者进行MTNR1B rs10830963基因分型。结果:80例患者完成8周随访,CG+GG基因型携带者空腹血糖(FPG)下降幅度、胰岛β细胞功能指数(HOMA-β)升高幅度均小于CC野生型携带者(P<0.05)。结论:MTNR1B rs10830963基因变异与瑞格列奈治疗T2DM的疗效相关,CC基因型携带者疗效更好。 AIM: To investigate the effects of MTNR1 B rs10830963 polymorphism on repaglinide response in patients with type 2 diabetes mellitus( T2DM). METHODS: Ninety newly-diagnosed patients with T2 DM were orally administrated 1 mg repaglinide three times per day for 8 consecutive weeks. The venous blood samples were collected at0 and 8th weekend. Anthropometric measurements and the parameters of glucose and lipid metabolism were determined before and after repaglinide treatment. The patients were enrolled to identify genotypes for MTNR1 B rs10830963 by the method of high resolution of melting curve. RESULTS: Eighty patients accomplished the 8-week follow-up visits.The reduction in FPG levels and increase in HOMA-β were lower in the carriers of genotype CG + GG at rs10830963 than those with the wild-type CC( P〈0. 05). CONCLUSION: The MTNR1 B rs10830963 polymorphism was associated with therapeutic efficacy of repaglinide in T2 DM patients. The CC homozygotes had a better effect than G allele carriers.
