摘要
目的探讨姜黄素介导光动力疗法(Photodynamic therapy,PDT)对裸鼠宫颈癌移植瘤模型的影响及其可能机制。方法将宫颈癌Me-180细胞悬液注入裸鼠皮下构建移植瘤模型,随机分为4组:模型组、PDT组、姜黄素组、姜黄素+PDT组,每组12只。各组治疗1 d后处死6只,剩余6只分别在第1、3、7、14、21天测量肿瘤体积。制备病理切片,观察各组HE病理改变、免疫组化。Westen-blot检测肿瘤组织中Notch-1蛋白的表达情况。结果模型组肿瘤体积逐渐增大,其余各组肿瘤体积在14 d内无增大趋势,随后逐渐增大。姜黄素+PDT组Notch-1蛋白表达与模型组比较,差异有统计学意义(P<0.01)。结论姜黄素介导的PDT能够有效抑制宫颈癌体外移植瘤的增长,其机制可能通过Notch-1信号通路发挥作用。
Objective To observe the effect of curcumin mediate Photodynamic therapy(PDT) on xenografted tumor of human cervical cancer and explore its mechanisms. Methods Human cervical cancer cells ( Me - 180)were implanted in the back of the Bal/bc nude mice. After filtration, all mice were randomly divided into 4 groups, including model group, PDT group, curcumin group, and curcumin + PDT group. There werel2 mice in each group. Six rats were executed one day after the treatment and the remaining 6 rats were left to measure the volume of the tumor on the 1s' ,3~ ,7th, 14th and 21st day. The pathological change of each group was observed by HE staining,immunohistochemistry. Western - blot was used to detect the expression of Notch - 1 protein. Results The tumor volume of model group increased gradually, while the other 3 groups had no changes in 14 days and then increased gradually. Compared with the model group, the Notch - 1 expression in the group of curcumin + PDT decreased significantly ( P 〈 0.01 ). Conclusion Curcumin me- diated PDT can effectively suppress the growth of cervical cancer transplanted tumor in vitro, and Notch - 1 signal pathway may play an important role in it.
