茵栀黄口服液对G6-PD酶活性下降大鼠溶血及红细胞膜ATP酶活性的影响 被引量:5

摘要 目的通过复制G6-PD酶活性下降大鼠模型,检测G6-PD酶活性及溶血相关指标,研究茵栀黄口服液是否会诱发或加重G6-PD酶活性下降大鼠红细胞溶血。方法按大鼠体重随机分为6组,正常组(记为A组),给予生理盐水,另外五组通过造模后分别处理:阴性对照组(给予生理盐水,记为B组),阳性对照组(给予伯氨喹啉,记为C组),茵栀黄大、中、小剂量组(给予口服液提取物,分别记为D、E、F组)。采用四唑氮蓝定量法(NBT)检测G6-PD酶活性。试剂盒测定血清间接胆红素含量、全血网织红细胞数量和血浆游离血红蛋白含量。超微量ATP酶测试盒测定红细胞膜Na+-K+-ATP和Ca2+-Mg2+-ATP酶的活性。结果 (1)E组大鼠G6-PD酶活性增强,与B组相比,差异有统计学意义(P<0.01),D、F组中大鼠G6-PD酶活性无明显改变;(2)与B组相比,伯氨喹啉可以明显升高血浆游离血红蛋白含量、全血网织红细胞数、血清间接胆红素含量,差异有统计学意义(P<0.01)。中小剂量茵栀黄口服液对上述指标均没有影响。而茵栀黄大剂量时可以明显升高血浆游离血红蛋白含量、全血网织红细胞数(P<0.01),血清间接胆红素也有升高趋势(P>0.05);3造模后大鼠红细胞膜Na+K+-ATP酶活性、Ca2+Mg2+-ATP酶活性均降低,E组可以增强大鼠红细胞膜Ca2+Mg2+-ATP酶活性,与B组比较,差异有统计学意义(P<0.05)。结论常用剂量的茵栀黄口服液可能不会诱发或加重鼠模型的溶血过程,相反,常用剂量可能增强鼠模型G6-PD及Ca2+Mg2+-ATP酶活性,对鼠模型红细胞膜可能发挥一定的保护作用。伯氨喹啉是已知的氧化型药物,能诱发鼠模型红细胞溶血,与文中研究得出的结论相吻合。 Objective To detect Glucose - 6 - Phosphate dehydrogenase ( G6 - PD ) enzyme activity and the related index of hemolysis ,to study weather Yin zhihuang oral liquid can induce or aggravate the he- molysis of red blood cells of rats with decreased G6 - PD enzyme activity. Methods Rats were divided into six groups randomly by weight, and one group was taken as norrnal group with normal saline, namely group A. Respective processing was applied on the other five groups after building models:negative control group( with normal saline, namely group B ) , positive control group ( with primaquine, namely group C ) , Yin zhihuang groups with high, medium and low dose (with oral liquid extract, namely group D, E, F). G6 -PD enzyme ac- tivity was detected by using nitrogen blue tetrazolium (NBT)method. The content of serum indirect bilirubin, plasma free hemoglobin and number of whole blood were also determined. At the same time, the enzymatic ac- tivity of Na · - K · - ATP and Ca2 · - Mg2 · - ATP on erythrocyte membrane were detected by ultramicro at- pase test kit. Results Compared with group B,the G6 - PD enzyme activity of group E enhanced greatly, and the difference was statistically significant( P 〈 0.01 ) ;For group D and F, there was no obvious changes in G6 -PD enzyme activity Compared with group B, pfimaquine could remarkably increase the content of serum indirect bilirubin, plasma free hemoglobin and number of whole blood, and the difference was statistically significant( P 〈 0.01 ) ,while indexes above -mentioned had no change for group E, F. Compared with group C, while Yin Zhihuang with large doses evidently increased the plasma free hemoglobin and blood reticulocyte count ( P 〈 0.01 ) , serum indirect bilirubin also raised ( P 〉 0.05 ) ; The enzymatic activity of Na - K - ATP and Ca2 - Mg2 - ATP on erythrocyte membrane both reduced after rat models established, but group E enhanced the enzymatic activity of Ca2 - Mg2 - ATP, compared with group B, and the difference was sta- tistically significant (P 〈 0.05). Conclusion Yin Zhihuang oral liquid may not induce or aggravate the he- molysis process of rat models, on the contrary, the enzymatic activity of G6 - PDand Ca2 - Mg2 - ATP may increase with common doses which can even play a protective role on erythrocyte membrane of rat models. As a kind of well- known oxidation drug,primaquine can induce erythrocyte hemolysis of rat models, which is consistent with the conclusion of this study.
