紫外线B照射对HeLa细胞中IEX-1mRNA表达的影响

摘要 目的:观察紫外线B(UVB)照射HeLa细胞后IEX-1 mRNA表达的变化。方法:常规培养人宫颈癌HeLa细胞,采用RT-PCR法检测剂量为0、2、4、8、16、32 m J/cm2的UVB照射30 s后HeLa细胞中IEX-1 mRNA的表达情况,同法检测剂量为32 m J/cm2的UVB照射后即刻及培养1、2、4、8、16 h时IEX-1 mRNA的表达情况。结果:随UVB剂量的升高,HeLa细胞IEX-1 mRNA的表达量呈升高的趋势(F=644.725,P<0.001)。32 m J/cm2的UVB照射HeLa细胞后即刻及培养1、2、4、8、16 h,IEX-1 mRNA的表达呈升高的趋势(F=56.249,P<0.001),培养4 h后IEX-1 mRNA表达水平达峰值。结论:HeLa细胞中IEX-1 mRNA的表达可被UVB迅速、短暂地诱导,且具有剂量依赖性。 Aim: To detect the effect of ultraviolet B(UVB) irradiation on the expression of IEX-1 mRNA in HeLa cells. Methods: HeLa cells were irradiated with 315 nm-UVB-ray at doses of 0,2,4,8,16,32 mJ/ cm2 for 30 s and then the level of IEX-1 mRNA expression was detected by RT-PCR. HeLa cells were irradiated with UVB-ray at the dose of 32 mJ/ cm2 for 30 s and the level of IEX-1 mRNA expression was detected by RT-PCR after being cultured for 0,1,2,4,8,16 h,respectively. Results: After being irradiated by different doses of UVB-ray, the expression of IEX-1 mRNA increased (F = 644. 725,P 〈 0. 001), especially at 32 mJ/ cm2 . After being irradiated by UVB-ray at the dose of 32 mJ/ cm2 , the ex-pression of IEX-1 mRNA increased at different time points(F = 56. 249,P 〈 0. 001), which was the highest at 4 h after ir-radiation. Conclusion: The expression of IEX-1 mRNA in HeLa cells could be quickly and transiently induced by UVB ir-radiation with a dose dependent trend.
