白酱感冒颗粒提取工艺及质量研究

摘要 目的:研究白酱感冒颗粒的提取工艺及其质量标准。方法:选用L9(34)进行正交设计实验,对白酱感冒颗粒的提取工艺进行筛选,以东莨菪内酯含量为指标,考察提取溶剂、提取时间、热回流速度、溶剂倍数4个因素;采用HPLC法测定东莨菪内酯的含量,色谱柱为依利特C18色谱柱(4.6mm×250mm,5μm);流动相为乙腈-水-冰醋酸(15∶85∶0.4);检测波长为343nm。结果:采用热回流提取技术加水煎煮2次,第一次提取2h,第二次提取1h,每次加水15倍,热回流速度为1.5L/h(以每kg药材计)提取效果最好;东莨菪内酯在2~12μg/m L线性关系良好(r=0.9998),平均回收率100.5%,RSD为1.69%。结论:该方法稳定可靠,适合工业化大生产。 Objective: To study the extraction process and quality standard of the Baijiang Ganmao Granule. Methods: Making an L9( 34) orthogonal experimental design,with the content of Scopoletin as the evaluation index and with the extraction solvent,extraction time,heat reflux rate,solvent multiples as the four factors. Finding out a optimization of the Baijiang Ganmao Granule extraction process; Using HPLC determination of Scopoletin,the column was Elite C18column( 4. 6mm × 250 mm,5μm); mobile phase of acetonitrile- water- acetic acid( 15∶ 85 ∶ 0. 4); detection wavelength of 343 nm. Results: Using the heat reflux extraction technology boil water 2 times,the first extract 2 hours and the second 1 hour,added 15 times of water every time,Heat reflux rate 1. 5L / h( per kg herbs) extracts the best; good linear indications showed as a results of the scopoletin being controlled within 2 ~ 12μg / m L( r =0. 9998),the average recovery was 100. 5%,RSD is 1. 69%. Conclusion: The method was stable and reliable,suitable for industrial production.
