青蒿-鳖甲药对配伍对MRL/lpr狼疮小鼠IL-6、IFN-γ分泌及狼疮性肾炎病理改变的影响 被引量:8

摘要 目的:观察青蒿-鳖甲药对对MRL/lpr狼疮模型小鼠相关免疫指标及狼疮性肾炎(lupus nephri-tis,LN)病理改变的影响,探索青蒿-鳖甲药对对系统性红斑狼疮(systemic lupus erythematosus,SLE)的治疗机理。方法:采用ELISA方法检测小鼠血清中的IL-6、IFN-γ含量,对肾脏组织进行HE常规染色及PAS、Masson特殊染色。结果:灌胃青篙-鳖甲药对水煎液后,低、高剂量组MRL/lpr小鼠血清中的IL-6水平显著低于正常对照组(P<0.05或P<0.01);高剂量组血清中的IFN-γ水平与模型组相比,显著下降(P<0.05);高剂量组肾脏组织HE、PAS、Masson染色病理改变差异显著。结论:青蒿-鳖甲药对能抑制MRL/lpr小鼠血清中IL-6的生成,减少小鼠血清中IFN-γ的分泌,能明显改善小鼠狼疮性肾炎的病理状态。 Objective: To investigate the effect of Qinghao- biejia group on auto- antibody production and pathologic change of lupus nephritis in MRL / lpr mice and the possible mechanism of qinghao- biejia group in treating systemic lupus erythematosus( SLE). Methods: IL- 6 and IFN- γ in serum of the MRL / lpr mice were detected by enzyme linked immunosorbent assay( ELISA). Renal tissue was stained by HE,PAS and Masson. Results: In the low and hight dose qinghao- biejia groups,as compared with the normal control group,the level of IL- 6 was significantly lower(P〈0. 05,P 0. 01); The level of IFN- γ was lower in the hight dose than in the model group(P〈0. 05); And renal pathological change was milder. Conclusion: qinghao- biejia group could inhibit the production of IL- 6 and secretion of IFN- γ and improve the pathologic lesion of lupus nephritis in MRL / lpr mice.
