摘要
目的分析微卫星不稳定性(MSI)在Ⅲ期结肠癌中导致的差异表达的肿瘤相关基因,以解释为何MSI与复发呈负相关。方法按特定筛选条件,搜寻NCBI公共基因表达数据库,使用R软件处理相关表达谱数据,包括统计学分析和差异基因分析。结果Ⅲ期结肠癌患者,在MSI未复发组与微卫星稳定性(MSS)复发组之间,通过差异基因表达分析,发现CAPN1、FLT3LG、SFN、ECT2、PTGS2、JUNB、ELF3、FOS、FOSB等基因在MSS复发组中高表达。PNAS4、ABCD3、CLDN8则在MSI未复发组中高表达。结论PNAS4、ABCD3和CLDN8这3个基因的高表达,可能是MSI患者复发率低的原因。而MSS患者复发率高,可能是因为癌基因JUNB、FOS的高表达,促使细胞癌变转移。
Objective To explore microsatellite instability (MSI)and differential expression of related genes in stage m colon cancer and explain the negative correlation between MSI and recurrence. Methods According to the specific conditions for screening, NCBI public gene expression database were R software applied for processing related expression data, including statistical analysis and differential gene analysis. Results For Stage m colon cancer patients, the analysis of differential gene expression showed that CAPNI, FLT3 LG, SFN, ECT2, PTGS2, JUNB, ELF3, FOS, and FOSB were highly expressed in micro- satellite stability(MSS) group (recurrence group). PNAS4, ABCD3, and CLDN8 were highly expressed in MSI group( no-recurrence group). Conclusion The high expression of PNAS4, ABCD3, and CLDN8 may be the reason for the low recurrence rate in patients with MSI. While the high recurrence rate in MSS patients may be contributed by the high expression of JUNB and FOS, which promotes cancerization and metastasis.
