汉坦病毒江西分离株全基因组序列分析 被引量:1

Analysis of the complete genome sequence of the Hantavirus isolates in Jiangxi

摘要 目的了解江西省汉坦病毒新分离株Jiangxi XinjianRn-07-2011的全基因组特征。方法设计特异性引物,分段扩增毒株的S、M、L全基因,对扩增产物进行序列测定,运用DNAStar、Mega 3.1软件对序列进行拼接和分析。结果全S、M、L基因分别含1 772个、3 651个、6 530个核苷酸,分别编码429个、1 133个、2 151个氨基酸,经序列比对和系统进化分析,该病毒属汉城型汉坦病毒(SEOV),S4亚型。S、M、L 3个基因与SEOV型病毒比较,核苷酸序列同源性分别为88.1%~98.0%、84.0%~96.1%、94.4%~95.9%;氨基酸序列同源性较高,分别为98.2%~100%、98.7%~99.5%、98.2~99.4%。结论测定了江西省新分离株Jiangxi XinjianRn-07-2011的全基因组序列,发现新分离株为SEOV,S4亚型。 Objective To understand the characteristics of the complete genome sequence of the Hantaviurs Jiangxi XinjianRn-07- 2011 new isolated strains in Jiangxi in 2011. Methods Design specific primers,and the complete S,M,L virus strains was amplified by segement and the amplified products were sequenced,the gene sequence were jointed and analyzed by DNAStar and Mega 3. 1 solftware. Results The complete S,M,L gene contained 1 772,3 651,6 530 nucleotides,encoding 429,1 133,2 151 amino,respectively. By sequence alignment and phylogenetic analysis,the virus belonged to seoul virus( SEOV),S4 subtype acids. Comparing S,M and L genes with SEOV virus. Nucleotide sequence homology was 88. 1%-98. 0%,84. 0%- 96. 1% and 94. 4%- 95. 9%,the level of homology in amino acid sequences was high and it was 98. 2%-100. 0%,98. 7%- 99. 5% and 98. 2- 99. 4%. Conclusion The complete gene sequence of Jiangxi XinjianRn- 07- 2011 new isolated strains was determined. The new isolates were SEOV,S4 subtype.
