摘要
[目的]探讨白花蛇舌草注射液对鼻咽癌CNE-2细胞体外增殖抑制作用及凋亡机制。[方法]MTT法检测白花蛇舌草注射液对鼻咽癌CNE-2细胞的抑制率,Annexin-V-FITC/PI双染法和Hoechst法检测细胞凋亡,Western blot法检测Bcl-2、Survivin、Bax、Caspase-3蛋白表达。[结果]不同浓度白花蛇舌草注射液对鼻咽癌CNE-2细胞具有抑制增殖作用,呈时间浓度依赖性(P<0.05)。8ml/L白花蛇舌草注射液能明显诱导CNE-2细胞凋亡,与对照组相比,具有统计学意义(P<0.05)。8ml/L白花蛇舌草注射液能诱导CNE-2细胞Bcl-2、Survivin蛋白表达降低、Bax及Caspase-3蛋白表达增加。[结论]白花蛇舌草注射液对鼻咽癌CNE-2细胞具有明显的体外增殖抑制作用,且呈时间浓度依赖性,其机制可能与诱导细胞凋亡,降低Bcl-2、Survivin蛋白表达,增加Bax及Caspase-3蛋白表达有关。
[Purpose] To investigate the anti-proliferation and apoptosis effects of Hedyotis diffusa in nasopharyngeal carcinoma CNE-2 cells in vitro and the mechanisms. [Methods] The inhibition effect was detected by MTT method. Apoptosis was detected by Annexin-V-FITC/PI and Hoechst.Bcl-2,Survivin,Bax and Caspase-3 expressions were detected by Western blot assay. [Results]The growth of CNE-2 cells was inhibited by Hedyotis diffusa in a time and dose manner(P〈0.05).Cell apoptosis was induced significantly by 8ml/L Hedyotis diffusa compare to the controls(P〈0.05). The Bcl-2 and Survivin proteins' expressions were decreased,and the Bax and Caspase-3 proteins' expressions were increased by 8ml/L Hedyotis diffusa. [Conclusion] Hedyotis diffusa might inhibit proliferation and induce apoptosis of CNE-2 cells in a time and dose manner,and the mechanism may be related to decreasing the levels of Bcl-2 and Survivin proteins and increasing the levels of Bax and caspase-3 proteins.
